    Assay conditions:

    Wells contained 50 mM Monobasic/Dibasic Phosphate Buffer pH 7.4, 20 μM H3, 1-21 (2MeK4), and varying amounts of KDM1 / LSD1 incubated for 30 minutes at room temperature. Followed by addition of 0.1 mM amplex red and 0.2 U/ml peroxidase.

    Fluorescence intensity measure at excitation 530 nm and emission 590 nm.

    Histone demethylase that demethylates both 'Lys-4' (H3K4me) and 'Lys-9' (H3K9me) of histone H3, thereby acting as a coactivator or a corepressor, depending on the context. Acts by oxidizing the substrate by FAD to generate the corresponding imine that is subsequently hydrolyzed. Acts as a corepressor by mediating demethylation of H3K4me, a specific tag for epigenetic transcriptional activation. Demethylates both mono- (H3K4me1) and di-methylated (H3K4me2) H3K4me. May play a role in the repression of neuronal genes. Alone, it is unable to demethylate H3K4me on nucleosomes and requires the presence of RCOR1/CoREST to achieve such activity. Also acts as a coactivator of androgen receptor (ANDR)-dependent transcription, by being recruited to ANDR target genes and mediating demethylation of H3K9me, a specific tag for epigenetic transcriptional repression. The presence of PRKCB in ANDR-containing complexes, which mediates phosphorylation of 'Thr-6' of histone H3 (H3T6ph), a specific tag that prevents demethylation H3K4me, prevents H3K4me demethylase activity of KDM1A. Demethylates di-methylated 'Lys-370' of p53/TP53 which prevents interaction of p53/TP53 with TP53BP1 and represses p53/TP53-mediated transcriptional activation. Demethylates and stabilizes the DNA methylase DNMT1. Required for gastrulation during embryogenesis.

Having reviewed the case, I would like to make some comments and raise a few more questions:

1. Both negative controls (Lane1) with two different antibodies are showing already a form ofnon-specific bands. Moreover, they are showing both the same pattern.

2. I checked the datasheet of the Histones, and it looks like that the sample you are using is actually a mixture of all the Histons H1 to H4. If you compare the molecular weight of the other Histones with the extra bands you are getting, it looks like both antibodies are actuallycross-reacting with Histons H2a to H4. However, I cannot explain why this effect becomes so prominent in Lane 2 to 4 in both blots.

3. I am concerned that the inhibitors seem to be without effect in blot one(lane 3 and 4) compared to lane 2. Therefore, I am actually wonderinghow you incubate the samples.

4. Could you please provide more information regarding theprotocol, especially how you incubated the samples?

5. Why are you adding DMSO?

6. The buffer of the KDM1 / LSD1 protein is actually protease free, and the ab17721 is picking nothing up:I am actually concerned that the protein is damaged rather than that the recombinant protein degrades Histone H3, what the reference you kindly send confirms.
