    ab96555 activity was measured by its ability to cleave a chromogenic peptide MMP3 substrate at room temperature.

    At a MMP3 concentration of 2.5 µg/ml, 50% cleavage was achieved at an incubation time of approximately 75 minutes.

Preparation and Storage

  • Stability and Storage

    Shipped at 4°C. Store at +4°C short term (1-2 weeks). Upon delivery aliquot. Store at -20°C or -80°C. Avoid freeze / thaw cycle. For long term storage it is recommended to add a carrier protein on reconstitution (0.1% HSA or BSA). Working aliquots stored with a carrier protein are stable for at least 3 months at -20°C to -80°C..

  • Reconstitution
    Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.5-1.0 mg/ml. Do not vortex.

    Can degrade fibronectin, laminin, gelatins of type I, III, IV, and V; collagens III, IV, X, and IX, and cartilage proteoglycans. Activates procollagenase.

The protocol for recombinant human MMP-3/MMP-2 bioactivity used is as follows:

Kit used is supplied by AnaSpec (Catolog#72095)


1. Dilute supplied colorimetric substrate 1:50 in assay buffer

2. Reconsitute MMP-3/MMP-2 sample with water (500ug/ml)

3. Mix 2ul MMP-3/MMP-2 and 400ul substrate solution

4. Take readings @412nm every 10 minutes for ˜90 minutes or until you have reached a steady reading.


According to the protocol booklet for this AnaSpec kit it states that the first step to activate the pro-MMPs is to indeed incubate with trypsin.

To our knowledge, this MMP3 protein has not been tested in a mouse system.

The amino acid sequence of the protein has approximately 78% identity to Mouse MMP3. Though not identical, it may still be capable of cleaving an endogenous mouse substrate, but we have no evidence for this.
