Sample: FITC conjugated Naloxone
Total protein input: 10nM

10nM of recombinant Human Mu Opioid Receptor protein (ab159029) was incubated with varying concentrations of FITC-conjugated Naloxone in 50mM Tris-HCl w/ 150mM NaCl for 30 minutes at room temperature. 50 µl reactions were then transferred to a 96-well ELISA plate previously coated with anti-GST antibody for an additional 1 hour incubation at room temperature. Wells were washed 1 time for 5 minutes with PBST (0.05% Tween20 in PBS), then 2 times for 5 minutes each with 50mM Tris-HCl w/ 150mM NaCl, and then imaged with a fluorescence multiwell plate reader. All experiments were performed in triplicate.

Conclusion: The recombinant Mu Opioid Receptor displayed slight binding activity at high concentrations of the tested compound, but the signal to background ratio was lower than human cell line expressed Mu Opioid Receptor.
