Functional Studies
Recombinant H3.1 was incubated with or without recombinant PADI2 (ab196406) or self produced GST-PADI4 in buffer conditions as described in Christophorou el al., 2014 Nature.
The N-terminal tail of Histone H3 has been shown to be citrullinated in vivo on various residues including Arginine 2. Our in vitro citrullination assay shows increased signal for H3R2-citrulline after incubation with recombinant PADI2 and 4 indicating the activity of the enzymes in vitro.
Loading:
Lane 1: 0.2ug of H3.1 (NEB)
Lane 2: 0.2ug of H3.1 (NEB) + 0.4 ul of PADI2 (ab196406 at 0.3ug/ml).
Lane 3: 0.2ug of H3.1 (NEB) + 500ng GST-PADI4
Blots were incubated o/n in anti-H3R2-Cit (ab176843) at 1:1000 in TBS with 5%BSA.
Secondary antibody ab6721 was used at 1:20000 dilution.
