    Growth factor that plays an essential role in the regulation of embryonic development, cell proliferation, cell migration, survival and chemotaxis. Potent mitogen for cells of mesenchymal origin. Required for normal proliferation and recruitment of pericytes and vascular smooth muscle cells in the central nervous system, skin, lung, heart and placenta. Required for normal blood vessel development, and for normal development of kidney glomeruli. Plays an important role in wound healing. Signaling is modulated by the formation of heterodimers with PDGFA.

    Expressed at high levels in the heart, brain (sustantia nigra), placenta and fetal kidney. Expressed at moderate levels in the brain (hippocampus), skeletal muscle, kidney and lung.

For ab73229, I was not able to receive any additional information for using the protein infunctional studies. As for ab9706, the procedure is as follows:

Procedure: (Previously reconstituted in water and stored at -20oC in 1XPBS+0.1%BSA)

1) Resuspend 3T3 cells in regular culture media with 10% calf serum. Transfer 100ml culture/well to assay plate (5,000 cells/well; passage # 7). Incubate overnight.

2) Complete media change with 100ml assay media containing 0.5% calf serum. Incubate overnight.

3) Complete media change with assay media.

4) Serial dilute h-PDGF-BB in assay media.

5) Transfer 100ml diluted cytokine to cells in assay plate. Volume of each well is 200ml, containing 0.5% calf serum and h-PDGF-BB as indicated below.

6) Incubate 42 hours.

7) Add 20ml of "other company" Substrate Cell Titer 96 Aqueous One Solution Reagent to each well.

8) Incubate 37oC and read at OD 490nm.


In direct comparison, ab9706 is a highly pure (>98%) recombinant human PDGF-BB (Platelet-Derived Growth Factor-BB), Expression host: E. coli, while ab73229 is arecombinant Full Length human PDGF BB is a homodimeric, glycosylated, polypeptide chain containing 109 (X 2) amino acids and having a molecular mass of 32 kDa, purified from Saccharomyces cerevisiae.

Yes, either HSA or BSA will be fine to use in the reconstitution of ab9706. We would also recommend a quick spin in a microcentrifuge prior to opening the vial to make sure the protein gets down to the bottom of the vial, then using sterile H2O and adding a carrier protein as mentioned if diluting below 0.1mg/ml (for example 0.1% BSA). If you have any further questions please do not hesitate to ask.

Both PDGF BB protein ab9706 (human) and ab51925 (mouse) have been assessed for activity by dose-dependent stimulation of the proliferation of Balb/c 3T3 cells with ED50 of 1.0-3.0 ng/ml and 2 ng/ml resepectively. They are both active. I am sorry that the information provided on the website has been confusing and thank you for bringing this to our attention. I will be amending this for future customers. ab9706 can be reconstitution with water, however a carrier protein is required if aliquotting under 0.1mg/ml. I hope this information is helpful to you.
