  • Function

    Mediates cAMP-dependent signaling triggered by receptor binding to GPCRs. PKA activation regulates diverse cellular processes such as cell proliferation, the cell cycle, differentiation and regulation of microtubule dynamics, chromatin condensation and decondensation, nuclear envelope disassembly and reassembly, as well as regulation of intracellular transport mechanisms and ion flux.
  • Tissue specificity

    Isoform 1 is most abundant in the brain, with low level expression in kidney. Isoform 2 is predominantly expressed in thymus, spleen and kidney. Isoform 3 and isoform 4 are only expressed in the brain.

    I have not been able to get the enzyme to phosphorylate a known substrate that I have made, i have tried several conditions as theproduct information sheet doesn't specify the best assay conditions. I am not sure whether the problem is with the enzyme or the substrate - do you sell/can you recommend a substrate that I can use as a control for PKA?

    I contacted the source of this protein and they would liketo receivedetailed information with regards to your protocol, substrate information, lot number.
    They need to have a thorough understanding of whathas beenperformed prior to make recommendations
    This information will also allow us to investigate this case internally and initiate additional testing where necessary. If the product was purchased in the last six months and is being used according to our Abpromise, we would be happy to replace or refund the protein.

    I can confirm that ab60837 was used in a kinase assay with CREBtide synthetic peptide substrate (KRREILSRRPSYR) , stock 1mg/ml.

    This is available for purchase from other sources.

    I attached the kinase assay protocol for your information.
