    Glycosylated, disulfide-linked homodimer of 1,976 amino acid residues with a calculated molecular weight of 226.2 kDa and therefore runs above the 200kDa marker by SDS-PAGE analysis under nonreducing conditions. 

    When run under reducing conditions, this protein migrates as three distinct bands that, due to glycosylation, run higher than expected at apparent molecular weights of approximately 120-130 kDa, 90-100 kDa, and 35-40 kDa.

  • Reconstitution
    Lot numbers beginning with GR3223939 should be reconstituted in 1xPBS to 0.1-0.25 mg/ml. Note: Allow the reconstituted vial to sit at room temperature for 1 hour before use. All other lot numbers should be in 20mM Tris, pH 8.5 to a concentration of 0.1-0.25 mg/ml. Note: Allow the reconstituted vial to sit at room temperature for 2 hours before use.   • Relevance

    One family of inhibitory axon guidance molecules is the semaphorins. The semaphorins include secreted, transmembrane, and GPI anchored extracellular molecules that are involved in regulating axon guidance by inhibiting axons from growing toward incorrect targets. Semaphorin 3A (Sema3A) may play a particularly interesting role in limiting axon regeneration since it is expressed in meningeal fibroblasts that invade the injured spinal cord and surround the glial scar. In addition, the Sema3A co receptors, Neuropilin 1 and Plexin A1, are expressed on axons that regenerate up to the injured region, but do not cross this Sema3A containing region. Thus, Sema3A and its co receptors may have important roles in regulating axon guidance during neuronal development and after neuronal injury.
