  • Function

    Converts the abundant, but inactive, zymogen plasminogen to plasmin by hydrolyzing a single Arg-Val bond in plasminogen. By controlling plasmin-mediated proteolysis, it plays an important role in tissue remodeling and degradation, in cell migration and many other physiopathological events. Play a direct role in facilitating neuronal migration.
  • Tissue specificity

    Synthesized in numerous tissues (including tumors) and secreted into most extracellular body fluids, such as plasma, uterine fluid, saliva, gingival crevicular fluid, tears, seminal fluid, and milk.
  • Involvement in disease

    Note=Increased activity of TPA results in increased fibrinolysis of fibrin blood clots that is associated with excessive bleeding. Defective release of TPA results in hypofibrinolysis that can lead to thrombosis or embolism.
  • Sequence similarities

    Belongs to the peptidase S1 family.
    Contains 1 EGF-like domain.

    Both FN1 and one of the kringle domains are required for binding to fibrin.

    The second kringle domain is implicated in binding to cytokeratin-8 and to the endothelial cell surface binding site.
  • Post-translational
    modifications

    The single chain, almost fully active enzyme, can be further processed into a two-chain fully active form by a cleavage after Arg-310 catalyzed by plasmin, tissue kallikrein or factor Xa.
    Differential cell-specific N-linked glycosylation gives rise to two glycoforms, type I (glycosylated at Asn-219) and type II (not glycosylated at Asn-219). The single chain type I glycoform is less readily converted into the two-chain form by plasmin, and the two-chain type I glycoform has a lower activity than the two-chain type II glycoform in the presence of fibrin.
    N-glycosylation of Asn-152; the bound oligomannosidic glycan is involved in the interaction with the mannose receptor.
    Characterization of O-linked glycan was studied in Bowes melanoma cell line.

    Mouse astrocytes uptake tPA-FITC

    tPA-FITC ab92679 was tested in an in vitro uptake assay performed on astrocytes isolated from P2-P3 mouse pups. The astrocytes were cultured in 4 well plates coated with PDL at a density of 25.000 cells/well in DMEM, 10% horse serum and 1% antibiotica (P/S).
    Before tPA application, the astrocytes were washed 2 times with pre-warmed DMEM to wash out the horse serum.
    After the washes, 140 nM tPA-FITC in DMEM was applied for 15 minutes at 37 degrees. Afterwards, the cells were washed 2 times with 1xPBS and subsequently fixed with 4% PFA at room temperature.
    For the ICC, after fixation, the cells were washed 2 times with 1xPBS, blocked with 1xPBS-Triton-X containing 10% NGS (goat serum) and incubated with a GFAP antibody (to visualise the astrocytes) for 1 hour at room temperature. Afterwards, the cells were washed 2 times with 1xPBS and the secondary antibody was applied together with DAPI to visualise the cell nuclei and incubated for 30 min at room temperature in the dark. After washing 3 times with 1xPBS, embedding medium was applied.
    For control conditions, some cells were incubated with 140 nM tPA-FITC in DMEM for 15 minutes at 4 degrees (cells do not uptake tPA in these conditions) and some cells with 140 nM Natural Human Serum Albumin protein-FITC (ab8030) in DMEM for 15 minutes at 37 degrees.

    Review text: Prior to using for fluoresecent confocal microscopy ab92679 (FITC-tPA) was tested for for fibrinolytic activity by measuring clot lysis using a turbidity assay. Clot lysis was observed with 20 pM ab92679.
    For observation of lysis by fluorescent confocal microscopy 75 nM ab92679 was used. A signal could be detected as expected at the lysis front.
