During development, promotes the survival and differentiation of selected neuronal populations of the peripheral and central nervous systems. Participates in axonal growth, pathfinding and in the modulation of dendritic growth and morphology. Major regulator of synaptic transmission and plasticity at adult synapses in many regions of the CNS. The versatility of BDNF is emphasized by its contribution to a range of adaptive neuronal responses including long-term potentiation (LTP), long-term depression (LTD), certain forms of short-term synaptic plasticity, as well as homeostatic regulation of intrinsic neuronal excitability.

Answer

I heard back from both labs that produce the BDNF and pro-BDNF proteins, and both said that the reconstitution instructions should be followed directly.
BDNF ab9794: reconstitute in water
pro BDNF ab138787: reconstitute in PBS

Answer

Thank you for contacting us.
The lab let me know the following:
The protein is lyophilized with no additives, which means that it is HPLC purified. Therefore, the only buffer that would be present at lyophilization would be water and 0.1% TFA (Triflouroacetic acid). This buffer is volatile, and will therefore evaporate out during the lyophilization process. Therefore, only a trace amount of this buffer would be present in the final formulation of the lyophilized protein.
The protein is > 98% pure by HPLC and SDS-PAGE.
Thus, we are not sure what is in the protein solution that could kill the birds, unless trace amounts of TFA could have such devastating effect.
We do however have not tested the protein in this way and also do not recommend using our products for in vivo studies.
I hope this information is of some help to you. Please do not hesitate to contact us if you need any more advice or information.

Answer

Question
Answer

Thank you very much for your call. Firstly, I would like to mention that we also have a PC12 cell lysate available that could be used as positive control for ab108383, as stated on the datasheet. I just point this out as a lysate may be of use as a control not only for this BDNF antibody, but for other antibodies in future experiments as well. This would be for example ab14883: Click here (or use the following: https://www.abcam.com/index.html?datasheet=14883). However, if you prefer the protein as positive control as discussed on the phone, it is absolutely fine to use ab9794 (Click here (or use the following: https://www.abcam.com/index.html?datasheet=9794).) As mentioned this morning, we do not currently have an image of ab9794 in SDS-PAGE/Western blot. Therefore, I can offer a discount off a future purchase if you buyab9794 now, use it in SDS-PAGE/Western blot and submit an image of your results to us in the form of an Abreview. The discount would be to the value of 1 free protein. It doesn’t matter whether the result is positive or negative, we would just really like to receive your feedback. Of course, if the results are negative you will be covered by our Abpromise guarantee and eligible for a refund or replacement. If you are interested in this offer, please follow these steps: 1. Reply to this e-mail to let me know that you would like to proceed and send an image of ab9794 in SDS-PAGE/Western blot. I will then send a discount code. This code must be issued before purchasing ab9794 so please wait for my reply before ordering. 2. Purchase ab9794 either by phone, fax, or online (www.abcam.com). 3. Use it in SDS-PAGE/Western blot. 4. Send us an image of the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: https://www.abcam.com/abreviews. 5. After the review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. The discount can be redeemed for any protein ordered and the discount code is valid for 4 months after issue. We are always pleased to obtain feedback about our products and any information is greatly appreciated! Furthermore, we are always happy to offer technical support and in the event that a product is not functioning in the applications cited on the product data sheet (and the problem has been reported within 6 months of purchase), we will be pleased to provide a credit note, free of charge replacement or refund. So if you need any advice or help with ab108383, please feel free to contact us. Please let me know if you have any questions about this offer and I would be happy to help you further. The Terms and Conditions of this offer can be found at: www.abcam.com/collaborationdiscount
