    Shipped at 4°C. After reconstitution store at -20ºC. Avoid freeze / thaw cycles.

    This product is an active protein and may elicit a biological response in vivo, handle with caution.

  • Reconstitution
    Reconstitute to a concentration of 0.1 mg/ml with ddH2O. Once reconstituted product is stable for at least 3 months when stored in working aliquots with a carrier protein at -200C.

  • Human embryonic stem cells (HUES4) were differentiated as monolayer cultures on matrigel coated dishes using a protocol described in Kroon et al, Nat Biotechnol. (2008). Inhibin beta A (ab50051) and Wnt3a (ab81484) were added to cultures to direct differentiation towards definitive endoderm. This image was taken 3 days after induction, where the definitive endoderm marker FOXA2 (ab40874 in green) is abundant. Dapi staining is shown in blue. Magnification: 200x. Negative controls of fluorescently conjugated secondary antibodies only showed no staining.

Functional Studies
This Wnt3a was used to treat mouse embryonic stem cells, which are quite responsive to Wnt3a.
We treated our cells for 24 hours with 200 ng/mL Wnt3a and then examined Wnt/beta-catenin target gene (e.g. Axin2) expression by qRT-PCR or Tcf/Lef activity by using a dual-luciferase reporter system (Super 8X-TOP/FOP-Flash).

The Wnt3a-mediated activation of the Wnt/beta-catenin pathway, as assessed by target gene activation or Tcf/Lef reporter activity, was not appreciably different from that observed when recombinant Wnt3a from other vendors was used.
