  • Function

    Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation.
  • Involvement in disease

    Genetic variations in TNF are a cause of susceptibility psoriatic arthritis (PSORAS) [MIM:607507]. PSORAS is an inflammatory, seronegative arthritis associated with psoriasis. It is a heterogeneous disorder ranging from a mild, non-destructive disease to a severe, progressive, erosive arthropathy. Five types of psoriatic arthritis have been defined: asymmetrical oligoarthritis characterized by primary involvement of the small joints of the fingers or toes; asymmetrical arthritis which involves the joints of the extremities; symmetrical polyarthritis characterized by a rheumatoidlike pattern that can involve hands, wrists, ankles, and feet; arthritis mutilans, which is a rare but deforming and destructive condition; arthritis of the sacroiliac joints and spine (psoriatic spondylitis).

Answer

For lot 851145: Centrifuge the vial prior to opening. Reconstitute in 200ul sterile water to a concentration of 0.1 mg/ml (or add 20ul for 1mg/ml). Do not vortex. This solution can be stored at 2-8oC for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20oC to -80oC.

Answer

I am terribly sorry about the delay replying to you regarding ab9756, I have had trouble contacting the source of this protein. I have just received confirmation that the protein gave only 1 band when tested in WB with the antibody ab9755. It is not likely that the second band is TNF beta but rather a non specific binding if you have used another antibody other than ab9755. Answer

Thank you very much for your patience, I have just heard back from the originator of these proteins. Ab9756: Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at 4C for 1 week or -20C for future use. Ab9970: Centrifuge the vial prior to opening. Reconstitute in 5mM Tris, pH 7.2 to a concentration of 0.1-1.0 mg/ml. This solution can then be diluted into other aqueous buffers and stored at 4C for 1 week or -20C for future use. Please let me know if you require any further information.
