      Streptavidin is a tetrameric protein purified from Streptomyces sp. that binds very tightly to the vitamin biotin with a Kd of ~ 10-14 mol/l. The high affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits.

    Gel used in the experiment is a 4-12% bis-tris invitrogen gel ran cold at 200V. The gel contains denatured and non-denatured lanes for ab78833 and ab156734. This shows that the protein is in the tetrameric form ‘out of the bottle’.

    If you are asking whether it’s detectable with a His tag antibody, then in principle it should be. If you are asking whether it’s the cleavable form, regrettably, it isn’t.

    I can confirm that the His tag is functional in this peptide for complexing with Nickel. The Met and Val in front of the His tag helps the His tag for proper configuration and has no impact on the protein’s functionality.
