Question
Answer

Thank you for taking the time to complete our questionnaire. The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality.

Reviewing this case, I would like to offer some suggestions to help optimize the results. I would also appreciate if you can confirm some further details:

1. I am sorry to confirm that according to the order details you have provided and our records, these items were shipped 30th December, 8 months ago. I am sorry this means they are regrettably no longer covered by our guarantee on this occasion. The guaranteeis for 6 months after purchase.

ab2627 Anti-RFC4 antibody

1. I can recommend to try lysing the sample in RIPA lysis buffer if you have not already done so. This should provide a suitable protein preparation. How were the nuclear extracts prepared?

2. Try a lower concentration of antibody to reduce non specific staining, try 1:4000.

3. We recommend to load 20 -30 ug of protein per lane of the gel to ensure it is not overloaded.

4. Try BSA rather than milk to block. Changing blocking agent can sometimes help to improve results.

5. The results look like a possible double band, I can recommend to ensure the sample is fully reduced and denatured.

6. Has the quality of the sample been assessed with a loading control?

7. I can recommend to include a no primary control to assess if there is any background staining from the secondary. Is the current vial of secondary antibody working well with other primary antibodies?

ab15340 Anti-RFC3 antibody

1. I can recommend to aliquot and store at the antibody at -20oC as stated on the datasheet.

We would suggest to store products as recommended on the datasheet as soon as possible after delivery. This will ensure it remains stable and that we can continue to provide the guarantee.

2. I can suggest to try increasing the antibody concentration to increase the signal. Try 1: 1000.

3. Has the quality of the sample been assessed with a loading control?

4. I can recommend to try lysing the sample in RIPA lysis buffer if you have not already done so. This should provide a suitable protein preparation. How were the nuclear extracts prepared?

5. Try BSA rather than milk to block. Changing blocking agent can sometimes help to improve results.

6. Is the current vial of secondary antibody working well with other primary antibodies? The concentration may need to be increased to help optimize the results.

ab94953 Anti-RFC1 antibody

1. Try a lower concentration of antibody to reduce the non specific binding. Try 1:2000. Incubate overnight 4oC.

2. Has the quality of the sample been assed with a loading control?

3. I can recommend to try lysing the sample in RIPA lysis buffer if you have not already done so. This should provide a suitable protein preparation. How were the nuclear extracts prepared?

4. Try BSA rather than milk to block. Changing blocking agent can sometimes help to improve results

5. Is the current vial of secondary antibody working well with other primary antibodies?

I can recommend to include a no primary control to assess if there is any background staining from the secondary.

6. The 'smile' effect on the band on the blot for this antibody indicate that the gel may have been run a little to quickly. Try a lower voltage for more time to ensure even bands.

I hope this information is helpful, thank you for your cooperation. Should the suggestions not improve the results, please do not hesitate to contact me again with the further requested details.

Answer

Thank you for taking the time to contact us. I am sorry to hear you have had difficulty obtaining satisfactory results from these antibodies.

I would like to reassure you that we monitor feedback closely on a weekly basis and we are not currently concerned about the general quality of these antibody or these batches. The antibodies are tested and covered by our 6 month guarantee forWB and human samples. RFC4 antibody (ab2627) is also tested in Saccharomyces cerevisiae. In the event that a product is not functioning in the tested applications and species cited on the product data sheet (and the problem has been reported within 6 months of purchase), we will be pleased to provide a credit note, free of charge replacement or refund.

In answer to your question, I am sorry to cnofirm that because we carry over 90,000 products, it isn't feasible for us to keep stock ofsmall sample sizes of our products to send to customers. However, I would like to investigate this particular case further for you, and also obtain more information for our quality monitoring records. In order to proceed with this, I have enclosed a technical questionnaire below. I would appreciate if you could complete this. It will help you put the information we require together very easily.

I would appreciate if you could also able to provide any images which would help us to assess the results.

Thank you for your time and cooperation. We look forward to receiving the completed questionaire.

Answer

Thank you for that extra information. I have arranged for a free of charge replacement with ab15340 to be sent. The order number of this is xxxxxxx. Unfortunately we did not have any of this antibody directly in stock, it will therefore most likely take until Friday to arrive as it needs to be shipped from our US office. I am sorry for any inconvenience this delay will cause. Thank you for offering to share the data you have obtained with ab57862. This will help greatly in investigating the case further.

Question
Answer

Thank you for calling us and for alerting us to the problem you are experiencing with one of our products. We take product complaints very seriously, and we investigate every product that we feel may not be performing correctly. As we discussed on the phone, I have looked into which antibody may be more appropriate for the experiments you are performing. ab15340 is a rabbit polyclonal to RFC3 which has been used in the Western blotting of endogenously expressed human protein, and would again be covered by the Abpromise for the experiment you are performing. As discussed over the phone, if you could provide me with the order number and lot number received I will be able to send this out to you free of charge, if you think it is appropriate for the experiments you are performing. It would be very helpful for me if you could provide some images of the experiments performed and maybe some protocol details. I have attached a questionnaire which I would appreciate if you could take the time to fill out (should only take 5 minutes). As I mentioned this will allow me to look into the case further allowing us to provide more complete details to future customers as well as initiating any additional testing required. Thank you for your cooperation and I look forward to your reply.
