    Regulates G protein-coupled receptor signaling cascades. Inhibits signal transduction by increasing the GTPase activity of G protein alpha subunits, thereby driving them into their inactive GDP-bound form (PubMed:11063746, PubMed:19478087). Plays a role in the regulation of blood pressure in response to signaling via G protein-coupled receptors and GNAQ. Plays a role in regulating the constriction and relaxation of vascular smooth muscle (By similarity). Binds EIF2B5 and blocks its activity, thereby inhibiting the translation of mRNA into protein (PubMed:19736320).

BATCH NUMBER 95043 ORDER NUMBER -- NOT SPECIFIED -- DESCRIPTION OF THE PROBLEM We're getting a band slightly higher than 50 kDa and a very faint band at approximately 35 kDa. Neither is close to the 25 kDa advertised. The band is very clean and there is no background. Could we be seeing RGS2 bound to its substrate? SAMPLE Rat pre-frontal cortex, homogonate PRIMARY ANTIBODY ab 14069 purchaced through Novus-Biologicals. Ab produced by Abcam. Ab 14069 is a chicken polyclonal to RGS2. Abtibody was diluted in blocker (see above) at 1/2000 and membrane was incubated for 1 hour at room temperature on a shaker. Following incubation the membrane was washed for 30 min. in 100 ml of washing solution (0.3%; 900 ml PBS with 2.7 ml Tween-20) SECONDARY ANTIBODY Secondary antibody: Anti-Chicken IgY (IgG) peroxidase conjugate from Sigma (product no. A 9046). Ab was developed in rabbit. Was diluted in blocker at 1:160,000 and applied for 1 hour at room temperature on a shaker, followed by two 15 minute washes in washing solution (see above). DETECTION METHOD ECL. Membrane was incubated in ECL solution for 5 min. ANTIBODY STORAGE CONDITIONS Stored upon arrival in a -80 degree freezer. SAMPLE PREPARATION Samples put into 10 microliters of sample buffer (sample buffer: 2 g SDS, 10mL glycerol, .005 g Bromophenol Blue, 0.6ml resolving gel buffer, fill with dH20 to 50 ml)with mercaptoethanol. Samples were then vortexed, boiled for 5 minutes and vortexed again prior to loading. AMOUNT OF PROTEIN LOADED 60 micrograms per lane ELECTROPHORESIS/GEL CONDITIONS 12% gel, Denaturing (SDS) discontinuous gel electrophoresis. Gel was run at 75V for 15 min. followed by 150V for 90 minutes. Proteins were then transferred to a nitrocellulose membrane. TRANSFER AND BLOCKING CONDITIONS Blocked on a shaker overnight in a cold room. Blocker (0.3%): 100ml PBS with 0.2g I-Block and 300 microliters of Tween-20. HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 2 HAVE YOU RUN A "NO PRIMARY" CONTROL? No DO YOU OBTAIN THE SAME RESULTS EVERY TIME? Yes WHAT STEPS HAVE YOU ALTERED? During the second run, we added an extra lane with double the amount of protein (approx. 120 micrograms). We did not see any difference. ADDITIONAL NOTES Since this is a polyclonal antibody could it be recognizing another member of the RGS family? Or alternatively could it be recognizing RGS2 with its substrate?

I'm sorry to hear you are having problems with ab14069. I would like to recommend to block for 1 hr with 5% milk and to incubate the primary antibody overnight in TBST at 4C, then wash in TBST several times before applying the secondary antibody. This antibody is raised against a full-length recombinant protein. It may recognize other related proteins too.
