Thank you for your reply. I am sorry that the replacement antibody that you received failed to work in western blot. I was wondering if with the replacement antibody you tried an overnight incubation of the primary antibody. I only ask, as I remember that during our previous troubleshooting emails, it was something we had discussed but not tried. Also, I know you were trying the antibody concentration of 1ug/ml, it may be worth increasing the concentration to 2ug/ml (1/500 dilution). It could be that you need to use the antibody at higher concentration and incubate for longer. I also remember that during our previous emails, you raised concerns about the presence of multiple bands in the western blot image shown for ab56560. Therefore I am concerned that if you were to use this antibody and you receive multiple bands, the antibody may still not be suitable.  I just wanted to mention this and see if you would still want to try ab56560. The alternative is that I could refund the original purchase for ab56560. I look forward to your reply.

Thank you for contacting Abcam. I am sorry that you are experiencing problems with ab83179 in western blotting. I have some questions/suggestions about the protocol that you are using. 1 - What concentration of ab83179 are you using? It has been tested at 1ug/ml, so depending on the concentration you are using, increasing the concentration could help. 2 - Incubating overnight at 4C is also recommended for antibodies that gave weak signal. 3 - Are you using 5% non-fat dried milk as your blocking agent? The antibody ab83179 is covered under our Abpromise and so is guaranteed to work in western blot on human samples. If we cannot resolve this issue then I would be happy to send you are replacement antibody or process a refund. I look forward to your reply.
