  • Function

    Subunit of mTORC2, which regulates cell growth and survival in response to hormonal signals. mTORC2 is activated by growth factors, but, in contrast to mTORC1, seems to be nutrient-insensitive. mTORC2 seems to function upstream of Rho GTPases to regulate the actin cytoskeleton, probably by activating one or more Rho-type guanine nucleotide exchange factors. mTORC2 promotes the serum-induced formation of stress-fibers or F-actin. mTORC2 plays a critical role in AKT1 'Ser-473' phosphorylation, which may facilitate the phosphorylation of the activation loop of AKT1 on 'Thr-308' by PDK1 which is a prerequisite for full activation. mTORC2 regulates the phosphorylation of SGK1 at 'Ser-422'. mTORC2 also modulates the phosphorylation of PRKCA on 'Ser-657'. Plays an essential role in embryonic growth and development.

I have contacted the laboratory to find out whether the immunogen sequence is similar to the Danio rerio sequence. The immunogen sequence is proprietary information, however the laboratory confirmed the following:

Sin1: At the site recognized by the antibody, human Sin1 ab71152 and the Zebrafish sequence (XP_003198622.1) are sufficiently similar to expect a probability of about 50% that the antibody ab71152 will bind Sin1 from Zebrafish.

Rictor: At the site recognized by the antibody, human Rictor ab70373 and the Zebrafish sequence (XP_001921907.1) are sufficiently divergent to expect the antibody ab70373 will not bind Rictor from Zebrafish.

I am very happy therefore to provide you with a discount code for the ab71152. Unfortunately I do not think that it would be the ab70373 is a good choice to test. I will be happy to look for possibility of crossreaction to Zebrafish of any of our other antibodies against rictor. Can you confirm please that you need the antibody for Western blot? Does it need to be a rabbit antibody?
