    The major RON transcript is translated into a glycosylated single chain precursor, cleaved into a 185 kDa heterodimer (p185Ron) of 35 (alpha) and 150 kDa (beta) disulfide-linked chains, before exposure at the cell surface.

    Mouse, Rat: We have preliminary internal testing data to indicate this antibody may not react with these species. Please contact us for more information.

      Overlay histogram showing THP1 cells stained with ab52927 (red line). The cells were fixed with methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab52927, 1/20 dilution) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-rabbit IgG (H+L) (ab96899) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was rabbit monoclonal IgG (1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a decreased signal in THP1 cells fixed with 4% paraformaldehyde (10 min) used under the same conditions.

      Please note that Abcam do not have any data for use of this antibody in non-fixed cells. We welcome any customer feedback.

    Answer

    Thank you for contacting Abcam regarding ab52927. I have confirmed that this antibody is an IgG1 isotype. Below, please find the IP protocol used: 1. Mix 200–300 μl of non-denatured cell lysate with appropriate dilutions of primary antibody 2. Incubate overnight at 4 °C with gentle rocking. 3. Add 25 μl of Protein G beads (50% bead slurry) and incubate 2 hours at 4 °C with gentle rocking. 4. Microcentrifuge for 30 seconds and discard the supernatant. Wash pellet 3 times with cold PBS. 5. Resuspend the pellet with 20 μl of 2x SDS reducing sample buffer containing β-mercaptoethanol (5%, v/v) and heat the sample at 95–100 °C for 3 minutes. 6. Briefly centrifuge the sample. 7. Load 16 μl of sample on to a SDS-PAGE gel or freeze the sample for later use (reboiled for 3 minutes prior loading onto the SDS-PAGE gel). 8. Perform Western blotting analysis using HRP-conjugated anti-rabbit IgG as secondary antibody  I hope this information is helpful.  Please do not hesitate to contact us if you have any additional questions.
