      I am very interested in this antibody. However, I have some questions that I could not find in you website. 1: Could you tell me the epitope of the antibody? 2: the protein of KIAA0460 is about 1440 amino acid, however the protein detected by this antibody is about 220kDa, How to explain? 3: Most important, how do you know this protein involved in DNA repair and DNA damage? I did not find any paper show this.

      Answer

      Thank you for your interest. The precise peptides used in the production of the antibodies ab10363 and ab10373 to KIAA0460 are proprietary. The antibody ab10363 is to a peptide that is encoded within the 5’ end of exon 11. The antibody ab10373 is to a peptide that is encoded within the 3’ end of exon 11. According to NCBI’s evidence viewer: (http://www.ncbi.nlm.nih.gov/sutils/evv.cgi?contig=NT_032962.5&gene=KIAA0460&lid=23248), Exon 11 encodes residues 529 to 1454 of BAA32305.2. The apparent MW on SDS-PAGE gels under the conditions in which the antibodies to KIAA0460 were tested is slightly larger than 220 kDa. Supporting evidence for this are: 1) both antibodies recognize a protein of the same Mr despite being generated to peptides that have nothing in common other than both are unique to KIAA0460 and 2) the antibodies were generated after KIAA0460 was identified as a protein that co-immunoprecipited with proteins that are established to be involved in repair of DNA damage. The gel slice, from which KIAA0460 was identified in this complex, was taken from the 220 to 240 Mr region of the gel. Owing to contractual obligations, the complex from which KIAA0460 was identified cannot be divulged prior to publication of these data.
