Epitope appears to be sensitive to PFA fixation. 15min at 4C in 4% PFA/PBS works best. Fixing for 2h at RTin 4%PFA/PBS works too. Longer fixations reduce the specific staining and result in dust like background which obscures the specifixc staining.
Section was counter stained with Hoechst nuclear stain for 10min at 22C in PBS ph7.4. Visible are the outer nuclear layer (top) and inner nuclear layer (bottom) of a adult mouse retina. Retionoid X receptor gamma staining was observed in the cone photoreceptors in the outer nuclear layer and also in most if not all cells of the inner nuclear layer.
