Target

  • Function

    Weakly binds calcium but binds zinc very tightly-distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites.

  • ab14849 at 1/3000 staining rat brain tissue sections by IHC-P. The tissue was formaldehyde fixed and a heat mediated antigen retrieval step was performed before incubation with the antibody. A biotinylated goat anti-mouse IgG was used as the secondary.

  • ab14849 staining S100 in human melanoma cultured cells by ICC (Immunocytochemistry). Cells were fixed with 4% paraformaldehyde in PBS, permeabilized with 0.2% Triton X-100 for 15 minutes and blocked for 30 minutes at 21°C. Samples were incubated with primary antibody (at concentrations listed above) for 15 hours at 4°C. A HRP-conjugated anti-mouse IgG polyclonal was used as the secondary antibody.

  • Interaction of several monoclonal antibodies with purified S100 protein (1 µg) from human brain in WB, after native gel electrophoresis using the Ornstein-Davis system.

    The doublet band appears at about 20-21 kD.

  • ab14849 staining S100 in the human Melanoma cell line 1205Lu xenograph tissue sections by IHC-P (Formaldehyde-fixed, Paraffin-embedded sections). Tissue samples were fixed with formaldehyde, permeabilized with 0.1% Triton X100 and blocked with a commercial block solution for 30 minutes at room temperature; antigen retrieval was by heat mediation in Tris EDTA at pH 9. The sample was incubated with primary antibody (1/300 in blocking solution) at 4°C for 18 hours. A biotin-conjugated goat polyclonal to mouse IgG (1/50) was used as secondary antibody.
