The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

    Target

    • Relevance

SARS (severe acute respiratory syndrome) is caused by a human coronavirus. Human coronaviruses are the major cause of upper respiratory tract illness, such as the common cold, in humans. Coronaviruses are positive-stranded RNA viruses, featuring the largest viral RNA genomes known to date (27-31 kb). The complete sequence of the SARS virus release the coronavirus contains 25 open reading frames. SARS E2 glycoprotein precursor is a 139kDa glycoprotein. It contains a superantigen between residues 690 through 1050 which has relationship to T-cell Receptor alpha-beta V chain protein.

    Question

ab39839  SARS E protein peptide    Could it be used with Anti-SARS E2 antibody [4A6C9] (ab31951) and Anti-SARS Nucleoprotein antibody [BDI186] (ab28169) to do sandwich assay? I want one antigen (SARS) and two monoclonal antibodies for the test of sars sandwich assay. Could you give me some suggestions ?Thank you!

    Answer

Thank you for your enquiry. I am sorry to confirm we do not have any SARS antibodies that are tested in sandwich ELISA (sELISA). SARS E2 antibody [4A6C9] (ab31951) and SARS Nucleoprotein antibody [BDI186] (ab28169) are tested in indirect ELISA only. I can suggest to consider trying one of them in an indirect ELISA method. Sandwich ELISA procedures can be difficult to optimise and tested match pair antibodies should be used. Any tested matched pairs would be noted on the datasheet. This ensures the antibodies are detecting different epitopes on the target protein so they do not interfere with the other antibody binding. Therefore, I am sorry we are unable to guarantee our antibodies in sandwich ELISA unless they have been specifically tested for sandwich ELISA. I am sorry we have no suitably tested antibodies for sELISA on this occasion. I can suggest having a look on the Biocompare website. They have an excellent antibody search facility which includes a lot of suppliers. Regrettably, I am also not able to confirm that the ab28169 is raised against the SARS nucleoprotein, so will not detect the ab39839 E2 protein peptide. However, the ab39839 peptide has been specifically used as the immunogen for ab31951 SARS E2 antibody [4A6C9], so these can be used together in indirect ELISA. I hope this will be helpful. Should you have any further questions, please do not hesitate to contact us. Good luck with your research.
