Target

  • Function
    Binds to DNA, at nuclear matrix- or scaffold-associated regions. Thought to recognize the sugar-phosphate structure of double-stranded DNA. Transcription factor controlling nuclear gene expression, by binding to matrix attachment regions (MARs) of DNA and inducing a local chromatin-loop remodeling. Acts as a docking site for several chromatin remodeling enzymes and also by recruiting corepressors (HDACs) or coactivators (HATs) directly to promoters and enhancers. Required for the initiation of the upper-layer neurons (UL1) specific genetic program and for the inactivation of deep-layer neurons (DL) and UL2 specific genes, probably by modulating BCL11B expression. Repressor of Ctip2 and regulatory determinant of corticocortical connections in the developing cerebral cortex. May play an important role in palate formation. Acts as a molecular node in a transcriptional network regulating skeletal development and osteoblast differentiation.

    High expression in adult brain, moderate expression in fetal brain, and weak expression in adult liver, kidney, and spinal cord and in select brain regions, including amygdala, corpus callosum, caudate nucleus, and hippocampus.
  • Involvement in disease
    Note=Chromosomal aberrations involving SATB2 are found in isolated cleft palate. Translocation t(2;7); translocation t(2;11).
    Defects in SATB2 are a cause of cleft palate isolated (CPI) [MIM:119540]. A congenital fissure of the soft and/or hard palate, due to faulty fusion. Isolated cleft palate is not associated with cleft lips. Some patients may manifest other craniofacial dysmorphic features, mental retardation, and osteoporosis.
    Note=A chromosomal aberration involving SATB2 is found in a patient with classical features of Toriello-Carey syndrome. Translocation t(2;14)(q33;q22).

  • ab51502 staining SATB2 in human fetal cortex tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).

    Tissue was fixed with paraformaldehyde and a heat mediated antigen retrieval step was performed using 10 mM citrate buffer pH 6.0. Samples were then blocked with 10% serum for 1 hour at 25°C followed by incubation with the primary antibody at a 1/100 dilution for 24 hours at 4°C. A Cy3®-conjugated horse anti-mouse polyclonal was used as secondary antibody at a 1/200 dilution. Counterstained with DAPI (blue).

  • ab51502 staining SATB2 (red) in a mouse cortex e17.5 cells by ICC/IF (Immunocytochemistry/immunofluorescence).

    Cells were fixed with paraformaldehyde, permeabilized with Triton X-100 and blocked with 3% donkey serum for 20 minutes at 22°C. Samples were incubated with primary antibody (1/10) for 90 minutes at 22°C. An Alexa Fluor®555-conjugated Donkey anti-mouse polyclonal (1/200) was used as the secondary antibody.

  • ab51502 staining cultured rat cortical neuron cells by ICC/IF.

    Cells were fixed in PFA, permeabilized in 0.1% triton and blocked with 10% BSA for 1 hour at 22°C.  The sample was then incubated with ab51502, diluted 1/25 for 2 hours at 22°C prior to incubation with a Cy3® conjugated donkey anti-mouse antibody, diluted 1/400.

Of the two rabbit monoclonals, ab92446 (SATB2) and ab92307 (SATB1), ab92446 is more likely to be specific for its target, SATB2, than ab92307 is for SATB1.

Alignments of the immunogens for each with the related protein returned 57% and 73% identity.

The immunogen for ab92446 anti-SATB2 was aligned with human SATB1 (UniProt Q01826) and the result was 4/7 (57%) over a 7 amino acid range.

The immunogen for ab92307 anti-SATB1 was aligned with human SATB2 (UniProt Q9UPW6) and the result was 11/15 (73%).

You mentioned in an earlier message that you were concerned that the antibodies ab51502 and ab92307 were not specific for the proteins SATB2 and SATB1, respectively:

The alignments of the immunogens with the other proteins do suggest that these antibodies may not have the specificity you hope for:

The ab92307 immunogen shares 73% identity (11/15) with human SATB2, Uniprot Q9UPW6.

The ab51502 immunogen shares 70% (72/103) identity with human SATB1, Uniprot Q01826.

Thank you for contacting us. The concentration of ab51502 is 0.1 mg/ml and should be used at a dilution of 1/100 for both IP and WB. Regarding ab92445, this is a tissue culture supernatant antibody so the concentration is not determined. Generally concentrations are only determined for purified antibodies as they are not relevant for other types that may contains proteins other than the protein of interest. This product should be used at a dilution of 1/10-1/100 for IP and 1/1000-1/10000 for WB. We do not test our antibodies side by side and cannot say which works "better" for a particular application but we do guarantee them both to work in IP and WB.
