Horseradish Peroxidase (HRP) has a high turnover rate that allows HRP secondary antibodies to generate strong signal in a short time span. HRP secondary antibodies are commonly used in western blot (WB), immunohistochemistry (IHC) and ELISA. Efficacy of HRP secondary antibodies is achieved by the optimal number of HRP molecules conjugated per antibody and specificity of the secondary antibody.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Goat Anti-Mouse IgG1 heavy chain (HRP) (ab97240).
A section of human colon formalin fixed paraffin embedded tissue section* was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH6) for 30mins. The section was incubated with anti-Histone H1.0 antibody (ab11080), 7.5µg/ml overnight at +4°C. A Goat Anti-Mouse IgG1 heavy chain (HRP) (ab97240) was used at 1/2000 dilution for 1hr at room temperature. The section was counterstained with haematoxylin and mounted with DPX.

Western blot - Goat Anti-Rabbit HRP (IgG H&L) (ab97051)
HeLa (Human epithelial carcinoma cell line) whole cell lysate (10 µg) was loaded in each lane. All lanes were incubated with anti-beta Actin antibody (ab8227) at 1 µg/ml as the primary antibody. Goat Anti-Rabbit HRP (IgG H&L) (ab97051) was used as the secondary antibody at 1/2000 dilution (lanes 1and 2), 1/10000 dilution (lanes 3 and 4) and 1/20000 dilution (lanes 5 and 6). The blot was developed using the ECL technique and performed under reducing conditions. Exposure time was 10 seconds.
