Horseradish Peroxidase (HRP) has a high turnover rate that allows HRP secondary antibodies to generate strong signal in a short time span. HRP secondary antibodies are commonly used in western blot (WB), immunohistochemistry (IHC) and ELISA. Efficacy of HRP secondary antibodies is achieved by the optimal number of HRP molecules conjugated per antibody and specificity of the secondary antibody.

“No nonspecific binding. No background. Clear bands.” (Researcher from Universidade do Porto, Portugal)

“Abcam’s anti-rabbit HRP is very reliable and economical to use. We ended up using 10x less than our old antibody from a different company. Since it is so robust, we started calling it the epic rabbit antibody to distinguish it from the old one we have in the lab.” (Researcher from University of Massachusetts Medical School, United States).

                      Goat anti-rabbit secondary antibody- ab205718

The substrates commonly used with HRP secondary antibodies fall into different categories including chromogenic (e.g. DAB, TMB, OPD), fluorogenic (e.g. ADHP) and chemiluminescent (e.g. ECL) substrates depending on whether they produce a colored, fluorimetric or luminescent derivative respectively.

Our HRP secondary antibodies have been extensively tested and optimized for diverse applications including WB, IHC and ELISA.

                      Abcam’s HRP secondaries advantages are:

                      • High sensitivity – detect low-abundant targets with high signal-to-noise ratios due to an optimal number of HRP molecules per antibody.
                      • Robustness – can be used at dilutions ranging from 1/2,000 to 1/20,000 and provide high specificity and minimal background.
                      • Stability – they are supplied in a liquid format that is ready-to-use and can be stored at 4°C.
                      • Flexibility – compatible with chromogenic, fluorogenic and chemiluminescent substrates.
                      • Cost-effective – use less conjugate per assay and perform more blots.

                      Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Goat Anti-Mouse IgG1 heavy chain (HRP) (ab97240).

A section of human colon formalin fixed paraffin embedded tissue section* was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH6) for 30mins. The section was incubated with anti-Histone H1.0 antibody (ab11080), 7.5µg/ml overnight at +4°C. A Goat Anti-Mouse IgG1 heavy chain (HRP) (ab97240) was used at 1/2000 dilution for 1hr at room temperature. The section was counterstained with haematoxylin and mounted with DPX.

                      Western blot - Goat Anti-Rabbit HRP (IgG H&L) (ab97051)

HeLa (Human epithelial carcinoma cell line) whole cell lysate (10 µg) was loaded in each lane. All lanes were incubated with anti-beta Actin antibody (ab8227) at 1 µg/ml as the primary antibody. Goat Anti-Rabbit HRP (IgG H&L) (ab97051) was used as the secondary antibody at 1/2000 dilution (lanes 1and 2), 1/10000 dilution (lanes 3 and 4) and 1/20000 dilution (lanes 5 and 6). The blot was developed using the ECL technique and performed under reducing conditions. Exposure time was 10 seconds.

                      * Preadsorbed (cross-adsorbed) antibodies go through an extra purification step to minimize species cross-reactivity and background. The process involves passaging of the antibody solution through a column containing immobilized serum proteins from potentially cross reactive species. Nonspecific cross-reactive secondary antibodies remain bound to the proteins in the column, while highly specific secondary antibodies are recovered from the flow through.
