Thank you for your enquiry. In testing this antibody, there is a lighter band at between 40 and 50 kD, in addition to the intense primary band at around 24 kD (the calculated molecular weight of SENP8). There are 5 cysteines in the SENP8 sequence (quite a few), so it is very important to make sure they are reduced and converted to the monomeric form. DTT (dithiothreitol) is approximately a 10x more powerful reducing agent than BME on a per molar basis, and is commonly used as a replacement when BME is not sufficient. It is important to make it fresh before use, as it loses potency within hours when exposed to air. Boiling a little longer could help, but we strongly advise using DTT. Also, SDS denatures proteins, but may not necessarily dissolve hydrophobic interactions that can increase migration rate. In these cases use of a non-ionic detergent that prevents hydrophobic nonspecific interactions is helpful, usually added at 0.01%. Nonidet P40 is one we often use; others in common usage include CHAPS, Triton-X, and Tween. Please do not hesitate to contact us again if you need anything further.
