    Serotonin ELISA kit is a competitive ELISA kit designed for the accurate quantitative measurement of Serotonin in Human Serum, Platelets, Plasma and Urine.


    A goat anti-rabbit IgG antibody has been precoated onto 96-well plates. Standards or test samples are added to the wells, along with an alkaline phosphatase (AP) conjugated- Serotonin antigen and a polyclonal rabbit antibody specific to Serotonin. After incubation the excess reagents are washed away. pNpp substrate is added and after a short incubation the enzyme reaction is stopped and the yellow color generated is read at 405 nm. The intensity of the yellow coloration is inversely proportional to the amount of Serotonin captured in the plate.

    Serotonin (5-hydroxytryptamine, 5-HT) is a monoamine found in the central nervous system, gastrointestinal tract, and blood with broad physiological functions as a neurotransmitter, in gastric motility, hemostasis, and cardiovascular integrity. Defects in serotonin signaling have been linked to a large number of complex behavioral disorders in humans, including anxiety and depression, autism, and eating disorders, and to neurodegenerative disorders such as Parkinson’s. Platelets serve as the major reservoir of serotonin in the bloodstream. When activated, platelets release serotonin into the bloodstream where it acts as a powerful vasoconstrictor. Treatment with Selective Serotonin Uptake Inhibitors (SSRIs) dramatically reduces platelet serotonin concentrations, and altered levels of serotonin in the circulatory system are implicated in such diverse conditions such as asthma, liver cirrhosis and carcinoid tumors.

    Dose-response curves from Human platelets and Human serum diluted into assay buffer were compared to the Serotonin standard curve. The parallel response indicates the standard effectively mimics the native protein.

Title: Quantitative measurement of serotonin in the cerebrospinal fluid (CSF)

Objective: quantitative measurement of serotonin in the cerebrospinal fluid (CSF) of patients with Parkinson's disease who underwent deep brain stimulation.
To assess the quality of the samples we performed prior quantification of total protein samples of cerebrospinal fluid through the Pierce's method.
It was possible to obtain levels around 0.30 ug / ul of total protein per sample. We use Serotonin Abcam kit for the detection of serotonin in the CSF samples and all controls and the standard curve functioned according to the specifications of the kit.
It was not possible to detect serotonin in the samples of these patients, suggesting that this level of serotonin in the cerebrospinal fluid is smaller than the detection capability of this kit.

Good kit to measure serotonin in mouse brain lysate

We have successfully used this ELISA kit to quantify serotonin levels in the frontal cortex of mice. 1mm thick sections of fresh tissue was extracted, kept on ice, and homogenised mechanically on the same day in 3ml of PBS. The calibration curve was easy to calculate manually on an Excel sheet and yielded excellent results, with both intra and inter-assay Coefficient of Variations below 30%. The one downside of this product is that little Assay Buffer is provided, thus limiting the possibility of multiple serial dilutions for every sample.

Good Kit For Measuring Serotonin in Rat Tissue Lysates and in Blood

We have used the ELISA kit and found that it works well on rat tissue lysates, CSF, and in blood samples. The only real draw back was that sometimes high amounts of protein are required, and can sometimes be problematic when working with small isolated brain regions, such as the the hippocampus or CSF samples, which have dilute protein concentrations.

Serotonin Elisa Kit with rat sample

I used the serotonin elisa kit with rat serum samples of tbi, ptsd, and control treatment groups. The kit gave decent results but I will say that the serotonin levels for my samples were significantly higher than the standard curve of the kit and also the standard error within treatment group was high. I would suggest anyone using this kit with rat serum samples should dilute the samples in half with the assay buffer. One thing that surprised me about this kit is that it calls for 100uL of serum/sample and they want you to do the samples in duplicate which is an enormous amount of serum in my opinion. On a positive note the directions were clear and easy to follow however you want to start this kit early in the day as there are two incubation steps that total three hours. On a side note the kit contains total activity, nonspecific binding, and maximum binding wells which were hard for me to label on my older plate reader which had none of these options when I went to set up my plate in the plate reader protocol. This may not be a problem for newer plate readers that were created for these more complicated elisa kits that require a 4-parameter logistic curve fit to determine concentration.

The kit was not specifically validated for cell culture supernatants but it is possible they may work for mouse assuming that there is enough for the kit to detect. Plus, Serotonin is not species specific (only samples containing endogenous
rabbit IgG would cause an issue). You would need to of course optimize dilution and be able to demonstrate dilutional linearity while falling onto the curve with the cell culture supernatant.

I wish to measure serotonin level in mouse brain. I know some people use ELISA kit for the measurement. However, many of these kits are formularised for use on biological fluid. Could you please advise if this kit is suitable for my application? Thanks.

The reason most kits are validated for fluids is because this marker is primarily expected to be in circulation/extracellular; therefore, the source of the kit informed me that they have not tested brain tissue let alone any other intracellular sample types.
As long as there is enough for the kit to detect it may work but sample handling/dilution optimization will be a necessary requirement of any customer especially when working with a non-validated sample type.
