    Serotonin transporter whose primary function in the central nervous system involves the regulation of serotonergic signaling via transport of serotonin molecules from the synaptic cleft back into the pre-synaptic terminal for re-utilization. Plays a key role in mediating regulation of the availability of serotonin to other receptors of serotonergic systems. Terminates the action of serotonin and recycles it in a sodium-dependent manner.

The images show different band sizes since the samples are different. One image shows a band at 75kda (as predicted). This image was generated with endogenous protein from mouse lung tissue lysate.

The other image was generated by overexpression of an ectopic Serotonin transporter protein and therefore the band is not depicting the predicted size.

I would like to assure you that ab102048 is tested and guaranteed with mouse samples for WB.

Une suggestion que je n'ai pas mentionnée dans mon précédent email est d'essayer d'utiliser 5% de BSA comme agent bloquant à la place du lait. Pour certains anticorps la BSA permet un signal plus intense et une réduction significative du bruit de fond, exemple avec l'antiGAPDH ab9385 https://www.abcam.com/ab9385.

Une utilisation de l'anticorps à une dilution au 1/1000 au lieu de 1/500peut peut-être également permettre d'obtenir un blot plus propre.

A notre connaissance, la réactivité contre les bivalves n'a pas encore été testée avec l'anticorps ab102048. Toutes les informations sur les cross-réactivités d'espèces sont spécifiées dans notre fiche technique, et sont mises à jour régulièrement.

- La séquence de la SERT est-elle connue pour la moule ou plus généralement pour les bivalves? Si oui, il serait intéressant de déterminer le degré d'homologie de cette protéine avec la SERT humaine et ainsi déterminer les chances que l'anticorps reconnaisse la protéine de moule. Je pense que l'absence de détection de la SERT chez la moule s'explique par la faible homologie de séquence entre ces 2 espèces phylogéniquement très éloignées.

- J'ai regardé vos blots et je pense que l'anticorps fonctionne bien et reconnaîtspécifiquement la SERT humaine dulysat de placenta.

- Pour le second controle, la SERT n'est pas exprimée dans le sang, ce qui explique l'absence de bande à 71kDa. Voir le lien suivant pour l'expression de la SERT: http://www.ncbi.nlm.nih.gov/UniGene/ESTProfileViewer.cgi?uglist=Hs.134662. La bande à 50 kDa est peut-être due au secondaire. Il serait intéressant de faire un blot avec le secondaire uniquement, sans primaire, afin de vérifier cette hypothèse.
