  • Function
    Actively transports glucose into cells by Na(+) cotransport with a Na(+) to glucose coupling ratio of 2:1. Efficient substrate transport in mammalian kidney is provided by the concerted action of a low affinity high capacity and a high affinity low capacity Na(+)/glucose cotransporter arranged in series along kidney proximal tubules.

  • ab14685 (2µg/ml) staining SGLT1 in human ileum using an automated system (DAKO Autostainer Plus). Using this protocol there is staining of the apical epithelium cells of the villi.
    Sections were rehydrated and antigen retrieved with the Dako 3 in 1 AR buffers EDTA pH 9.0 in a DAKO PT link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 mins. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS) then incubated with primary antibody for 20 min and detected with Dako envision flex amplification kit for 30 minutes. Colorimetric detection was completed with Diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that, for manual staining, optimization of primary antibody concentration and incubation time is recommended. Signal amplification may be required.

Thank you for your inquiry. I can confirm that rat SGLT1 has a predicted  molecular weight of ca. 73kDa. Molecular weights on gel are always only a rough estimate since the gel and buffer conditions can influence the resulting molecular weight of a protein dramatically. It is know in this case that also the percentage of the gel has an influence on the molecular weight of SGLT1. I hope i could reassure your customer.
