Target

  • Function

Seems to be an adapter protein in the postsynaptic density (PSD) of excitatory synapses that interconnects receptors of the postsynaptic membrane including NMDA-type and metabotropic glutamate receptors via complexes with GKAP/PSD-95 and Homer, respectively, and the actin-based cytoskeleton. Plays a role in the structural and functional organization of the dendritic spine and synaptic junction.
  • Tissue specificity

Expressed in brain particularly in the amygdala, hippocampus, substantia nigra and thalamus. Isoform 2 seems to be expressed ubiquitously.
  • Sequence similarities

    Belongs to the SHANK family.
    Contains 6 ANK repeats.
    Contains 1 PDZ (DHR) domain.
Contains 1 SAM (sterile alpha motif) domain.
    Contains 1 SH3 domain.
  • Cellular localization

Cytoplasm. Cell junction > synapse > postsynaptic cell membrane > postsynaptic density. Cell junction > synapse. Postsynaptic density of neuronal cells.

  • Western blot detection of Shank1 in Rat brain membrane lysates using ab94576 at 1:1000 dilution.
  • paraffin-embedded, bouin's-fixed mouse backskin stained for SHANK1 using ab94576 at 1/100 dilution in immunohistochemical analysis. FITC Goat Anti-Mouse (green) at 1/50 dilution was used as the secondary antibody. Filaggrinlike staining (upper layer aggregations of staining) is observed.

  • Overlay histogram showing SH-SH5Y cells stained with ab94576 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab94576, 1μg/1x106 cells) for 30 min at 22°C. The secondary antibody used was Alexa Fluor® 488 goat anti-mouse IgG (H&L) (ab150113) at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 1μg/1x106 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. This antibody gave a positive signal in SH-SY5Y cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

Answer

Thank you for contacting us. Because we carry over 100,000 products, it isn't feasible for us to keep small sample sizes of our products.

We are happy to reassure our customers that all of our products are covered by our Abpromise, which guarantees that the product will work in the applications and species specified on the datasheet, or we will offer a replacement, credit, or refund within 6 months of purchase.

If the product is to be used in an untested species or application, as in your case you would be eligible for our AbTrial program because the antibody has not yet been tested in ELISA. To find out more information about this program view the following link:

www.abcam.com/abtrial

Otherwise, we like to encourage all of our customers to submit an Abreview via the online product datasheet. We always appreciate customer feedback, whether positive or negative, and we make all product information available to researchers. Plus, each Abreview earns Abpoints that can be used for discounts on future purchases or rewards such as Amazon.com gift certificates.

To find out more about our Abreview system, please see the following link:

Answer

Thank you very much for your interest our product and for your patience.

To our knowledge, ab94576 has not been tested in ELISA. I searched for another ELISA suitable SHANK1 antibody and regrettably could not find any that has been tested in ELISA.

For this reason I would like to offer you to participate in our AbTrial programme by testing the ab94576 in ELISA with a teted species. By participating in our AbTrial program you can use our products in an untested application or species without financial risk.

Simply follow these easy steps below to apply for our AbTrial Program:

1. Reply to this email, letting me know you are interested in testing this product.

2. I will email you an inactive personal discount code for the value of the product.

3. Purchase and test the product at the regular price.

4. Submit your results, including your discount code in the additional notes section of your Abreview.

5. Once the Abreview is submitted, the discount code will become active.

6. Apply your discount code on your next order to receive that value off.

Please let me know if you have any questions about this offer and I would be happy to help you further.
