  • Function
    Acts as an endocytic receptor mediating clathrin dependent endocytosis. Macrophage-restricted adhesion molecule that mediates sialic-acid dependent binding to lymphocytes, including granulocytes, monocytes, natural killer cells, B-cells and CD8 T-cells. Preferentially binds to alpha-2,3-linked sialic acid (By similarity). Binds to SPN/CD43 on T-cells (By similarity). May play a role in hemopoiesis.
  • Tissue specificity
    Expressed by macrophages in various tissues. High levels are found in spleen, lymph node, perivascular macrophages in brain and lower levels in bone marrow, liver Kupffer cells and lamina propria of colon and lung. Also expressed by inflammatory macrophages in rheumatoid arthritis.

  • ab18619 staining human peripheral blood mononuclear cells by flow cytometry. The cells were prepared in FACS buffer (PBS, 0.5% FCS, 0.5M EDTA) and fixed in PFA without undergoing permeabilization. Cells were gated on lymphocyte and monocyte populations and incubated with primary antibody (1/25) for 20 minutes at 4°C. The secondary antibody used was Phycoerythrin-conjugated rat polyclonal to mouse Ig, diluted 1/100. The PBMC cells were almost exclusively CD14+ monocytic cells.

  • ICC/IF image of ab18619 stained MCF7 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab18619, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

One reference listed in the 'Specific references' section does relate use of ab18619 in IHC. This is the Hartnell et al paper. They do not describe the use of antigen retrieval.

I would suggest trying no antigen retrieval along with a citrate buffer pH6 antigen retrieval and compare the results to see which gives the best staining.
