The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 1 - 2 µg/ml. Predicted molecular weight: 43 , 38 kDa.
IP Use at an assay dependent dilution. Use at 2 - 5 µg.
ICC/IF Use a concentration of 5 - 10 µg/ml.

  • Function

NAD-dependent protein deacetylase, which deacetylates the 'Lys-40' of alpha-tubulin. Involved in the control of mitotic exit in the cell cycle, probably via its role in the regulation of cytoskeleton.
  • Tissue specificity

Widely expressed. Highly expressed in heart, brain and skeletal muscle, while it is weakly expressed in placenta and lung. Down-regulated in many gliomas suggesting that it may act as a tumor suppressor gene in human gliomas possibly through the regulation of microtubule network.
  • Sequence similarities

    Belongs to the sirtuin family.
    Contains 1 deacetylase sirtuin-type domain.
  • Developmental stage

Peaks during mitosis. After mitosis, it is probably degraded by the 26S proteasome.
  • Post-translational

Phosphorylated at the G2/M transition of the cell cycle.
  • Cellular localization

Cytoplasm > cytoskeleton. Colocalizes with microtubules.

  • ab67299 staining SIRT2 in pure rat Schwann cells by ICC/IF (Immunocytochemistry/immunofluorescence) after resveratol (RSV) treatment. Cells were fixed with 4% PFA blocked with 10% Goat serum/ 0.1% Triton x-100/ 0.1% BSA in PBS for  60 minutes at 21°C followed by 10% Goat serum/ 0.5% Triton X-100/ 0.01% BSA in PBS for 15 minutes at 21°C. Samples were incubated with primary antibody (1/100 in PBS + 10% goat serum) overnight at 21°C. An  Alexa Fluor®488-conjugated goat anti-mouse IgG polyclonal (1/400) was used as the secondary antibody. Stimulation  of rSCs with RSV led to an increase of SIRT2 expression in densitometry analysis.

Thank you for contacting us. SIRT6 The human, mouse and rat proteins are homologous in N terminal and central region of protein. So the antibodies available to these regions will most likely cross-react. ab62739, cross reacts with human, mouse and rat. Not yet tested in Immunohistochemistry.  ab105391, cross reacts with human, mouse and rat. Not yet tested in Immunohistochemistry. These antibodies are not yet tested in Immunohistochemistry. However this does not mean that these are not suitable for IHC it only mean we do not have tested data to confirm their use. If you are interested in testing one of these in IHC then let me know I will send you the Abreview discount code;  which states we will offer a next free antibody in exchange of an antibody review.    SIRT1 ab32441 and ab32424 These antibodies are tested in IHC so are fully guaranteed to work in IHC. The mouse and rat species is predicted which is based on sequence homology of immunogen between these species. If you are interested in using these antibodies in mouse and rat then let me know I will provide you the Abreview discount code which will give you your next purchase of an antibody free of charge. ab75435 and ab110304 does cross react with human, mouse and rat however not tested in IHC therefore these are eligible for Abreview discount programme.   SIRT2 ab115681 is tested in human and mouse and in IHC-P. ab67299 and ab75436 are tested for cross reactivity in human, mouse and rat however not tested in IHC. These products are also eligible for Abreview discount programme. I also have attached a spreadsheet of all anti SIRT6, SIRT2 and SIRT1 antibodies with Immunogen sequence. Please have a look and decide which one best suits you. For information about publications and images please check the datasheets of individual antibodies. I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
