Target

  • Function

Translocates a broad array of organic cations with various structures and molecular weights including the model compounds 1-methyl-4-phenylpyridinium (MPP), tetraethylammonium (TEA), N-1-methylnicotinamide (NMN), 4-(4-(dimethylamino)styryl)-N-methylpyridinium (ASP), the endogenous compounds choline, guanidine, histamine, epinephrine, adrenaline, noradrenaline and dopamine, and the drugs quinine, and metformin. The transport of organic cations is inhibited by a broad array of compounds like tetramethylammonium (TMA), cocaine, lidocaine, NMDA receptor antagonists, atropine, prazosin, cimetidine, TEA and NMN, guanidine, cimetidine, choline, procainamide, quinine, tetrabutylammonium, and tetrapentylammonium. Translocates organic cations in an electrogenic and pH-independent manner. Translocates organic cations across the plasma membrane in both directions. Transports the polyamines spermine and spermidine. Transports pramipexole across the basolateral membrane of the proximal tubular epithelial cells. The choline transport is activated by MMTS. Regulated by various intracellular signaling pathways including inhibition by protein kinase A activation, and endogenously activation by the calmodulin complex, the calmodulin-dependent kinase II and LCK tyrosine kinase.
  • Tissue specificity

Widely expressed with high level in liver. Isoform 1 and isoform 2 are expressed in liver. Isoform 1, isoform 2, isoform 3 and isoform 4 are expressed in glial cell lines.
  • Sequence similarities

Belongs to the major facilitator superfamily. Organic cation transporter family.
  • Post-translational
    modifications

    Phosphorylated.
  • Cellular localization

    Basolateral cell membrane.

Answer

Thank you for your inquiry and your patience with this reply.
I heard back from the lab regarding ab55916. The antibody is a short 14 a.a. peptide located in this sequence: LATNRSHLPL GPCQDGWVYD TPGSSIVTEF NLVCADSWKL DLFQSCLNAG. This sequence is fused to KLH for animal immunization, and then fused to agarose beads for the purification process. You can further purify this antibody if you would like.
For ab107246, we may be able to provide this antibody purified, but it may require a minimum order of at least 10 vials. If you are interested in a custom bulk order, please email our sales team at sales@abcam.com.
I hope this information helps. Please contact us with any other questions.

Answer

Thank you your reply and for keeping me updated. I am sorry to hear that the results with ab105049 are still not as expected.
I would be happy to send a new vial of this antibody, however we only have the same batch in stock. Would you like to try a new vial from this batch? If so, I will send it promptly. Alternatively I can issue a credit or refund for your original purchase.
I look forward to hearing from you. Please let me know if you have any questions or if there is anything else that we can do for you.

Answer

Thank you for your reply and for sending the additional information and the images.
For antigen retrieval prior to using ab55916, the lab microwaved the tissue in pH6 0.01 M citrate buffer for 10 minutes at ˜100C, and then the section were cooled to room temperature for 30-60 minutes. For retrieval prior to using ab105049, the tissue was microwaved in 1 mM pH8 EDTA for 10 minutes and then again cooled for around 20 minutes. Is this similar to the treatments that you have tried? The rest of your protocol seems to be very similar to the lab protocols.
Based on information provided by the Human Protein Atlas, SLC47A1 should be showing as cytoplasmic/membranous staining mostly strongly in tubule cells of the kidney-
http://www.proteinatlas.org/ENSG00000142494/normal/kidney
The SLC22A1 should also be cytoplasmic/membranous, though this protein seems to be expressed more weakly and uniformly in the kidney-
http://www.proteinatlas.org/ENSG00000175003/normal/kidney
It's difficult to really see the localization of the staining in the images that were sent, so you probably have a better idea of whether your staining is specific or not. The SLC22A1 staining seems to be similar, but the SLC47A1 does not. You may want to try antigen retrieval methods as described above, however if you've already tried similar methods then I'd be happy to send new vials of these antibodies for you to try.
I look forward to hearing from you. Please let me know if you would like to try fresh vials of antibodies and I will send this out promptly. Let me know if you have any questions or if there is anything else that we can do for you.

Answer

Thank you for contacting us and for letting us know about the trouble with these antibodies in IHC-P.
Thank you also for sending the protocol information. I do have a couple of additional questions in order to better understand the situation:
1) Do you have your order number or lot number?
2) How has the antibody been stored?
3) Which concentrations were tried (higher or lower end of the recommended concentrations)?
4) What were the results? Is there high background, no staining at all, or staining the incorrect localization? Would it be possible to send images?
5) Are these antibodies being used together on the same tissue, or separately?
6) Were there any differences in the results when the protocol was altered (i.e. using different antigen retrieval methods or blocking solution)?
I look forward to hearing from you and resolving this promptly. We do guarantee all of our products to work as stated on the datasheets for up to 6 months after purchase, so I would be happy to send replacements or issue credits or refunds if we can not improve the results.
Please let me know if you have any questions or if there is anything else that we can do for you.
