Application Abreviews Notes
WB Use a concentration of 2.5 µg/ml. Detects a band of approximately 61 kDa (predicted molecular weight: 61 kDa).Can be blocked with Human SLC22A1/OCT1 peptide (ab111655). Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
IHC-P Use a concentration of 4 - 8 µg/ml.


  • Function

    Translocates a broad array of organic cations with various structures and molecular weights including the model compounds 1-methyl-4-phenylpyridinium (MPP), tetraethylammonium (TEA), N-1-methylnicotinamide (NMN), 4-(4-(dimethylamino)styryl)-N-methylpyridinium (ASP), the endogenous compounds choline, guanidine, histamine, epinephrine, adrenaline, noradrenaline and dopamine, and the drugs quinine, and metformin. The transport of organic cations is inhibited by a broad array of compounds like tetramethylammonium (TMA), cocaine, lidocaine, NMDA receptor antagonists, atropine, prazosin, cimetidine, TEA and NMN, guanidine, cimetidine, choline, procainamide, quinine, tetrabutylammonium, and tetrapentylammonium. Translocates organic cations in an electrogenic and pH-independent manner. Translocates organic cations across the plasma membrane in both directions. Transports the polyamines spermine and spermidine. Transports pramipexole across the basolateral membrane of the proximal tubular epithelial cells. The choline transport is activated by MMTS. Regulated by various intracellular signaling pathways including inhibition by protein kinase A activation, and endogenously activation by the calmodulin complex, the calmodulin-dependent kinase II and LCK tyrosine kinase.

I heard back from the lab regarding ab55916. The antibody is a short 14 a.a. peptide located in this sequence: LATNRSHLPL GPCQDGWVYD TPGSSIVTEF NLVCADSWKL DLFQSCLNAG. This sequence is fused to KLH for animal immunization, and then fused to agarose beads for the purification process. You can further purify this antibody if you would like.

For antigen retrieval prior to using ab55916, the lab microwaved the tissue in pH6 0.01 M citrate buffer for 10 minutes at ˜100C, and then the section were cooled to room temperature for 30-60 minutes. For retrieval prior to using ab105049, the tissue was microwaved in 1 mM pH8 EDTA for 10 minutes and then again cooled for around 20 minutes. Is this similar to the treatments that you have tried? The rest of your protocol seems to be very similar to the lab protocols.
Based on information provided by the Human Protein Atlas, SLC47A1 should be showing as cytoplasmic/membranous staining mostly strongly in tubule cells of the kidney-
The SLC22A1 should also be cytoplasmic/membranous, though this protein seems to be expressed more weakly and uniformly in the kidney-
It's difficult to really see the localization of the staining in the images that were sent, so you probably have a better idea of whether your staining is specific or not. The SLC22A1 staining seems to be similar, but the SLC47A1 does not. You may want to try antigen retrieval methods as described above, however if you've already tried similar methods then I'd be happy to send new vials of these antibodies for you to try.

1) Do you have your order number or lot number?
2) How has the antibody been stored?
3) Which concentrations were tried (higher or lower end of the recommended concentrations)?
4) What were the results? Is there high background, no staining at all, or staining the incorrect localization? Would it be possible to send images?
5) Are these antibodies being used together on the same tissue, or separately?
6) Were there any differences in the results when the protocol was altered (i.e. using different antigen retrieval methods or blocking solution)?
