  • ab78066, at 20µg/ml, staining SLC40A1 in human skeletal muscle by Immunohistochemistry using formalin-fixed, paraffin-embedded tissue.
    A heat induced antigen retrieval step was performed using pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen.
  • ICC/IF image of ab78066 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab78066, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

I am sorry that you have been experiencing difficulties with this antibody in IHC of frozen sections. Unfortunately, this antibody has not been tested in IHC of frozen sections and it is therefore possible that it is not suitable for this application.

I am hoping you would be willing to provide some additional information about how you used the antibody in your experiments. If you could reply with the details of your sample preparation and IHC protocol, I may be able to offer some protocol advice to help you troubleshoot. Since we have not validated ab78066 for IHC on frozen sections, we are not able to guarantee the product per our Abpromise.
