    Acts as an electrogenic sodium (Na(+)) and chloride (Cl-)-dependent sodium-coupled solute transporter, including transport of monocarboxylates (short-chain fatty acids including L-lactate, D-lactate, pyruvate, acetate, propionate, valerate and butyrate), lactate, mocarboxylate drugs (nicotinate, benzoate, salicylate and 5-aminosalicylate) and ketone bodies (beta-D-hydroxybutyrate, acetoacetate and alpha-ketoisocaproate), with a Na(+):substrate stoichiometry of between 4:1 and 2:1. Catalyzes passive carrier mediated diffusion of iodide. Mediates iodide transport from the thyrocyte into the colloid lumen through the apical membrane. May be responsible for the absorption of D-lactate and monocarboxylate drugs from the intestinal tract. Acts as a tumor suppressor, suppressing colony formation in colon cancer, prostate cancer and glioma cell lines. May play a critical role in the entry of L-lactate and ketone bodies into neurons by a process driven by an electrochemical Na(+) gradient and hence contribute to the maintenance of the energy status and function of neurons.
  • Tissue specificity

    Expressed in normal thyroid, localized at the apical pole of thyroid cells facing the colloid lumen, but expression profoundly decreased in thyroid carcinomas. Expressed in normal colon but absent in colon aberrant crypt foci and colon cancers. Present in normal kidney cortex, brain, prostate, gastric mucosa and breast tissue but was significantly down-regulated in primary gliomas, gastric cancer, prostate tumors and breast tumors.

In order to help you further could you provide me with the order number and lot number of the antibody? xxx xxx I would also like to get some further information for our records about the protocol used for your blotting. "Standard BioRad reagents.  . Samples denatured at 37oC for 30 min as recommended for membrane proteins such as SLC5A8" Could you please send me information as to the concentration of the antibody that you have used and for how long?   "We used 1:500 dilution (original 1mg/ml) and incubated at 4C for O/N (16h) ." What your blocking solution was and if you tried different blocks? "Our blocking buffer is 5% nonfat dry milk in TBST. " What detection method did you use?  "PVDF membrane after secondary antibody was incubated in Pierce Femto reagent for 2-5min and then detected with chemiluminescent imaging (Alpha Innotech Imager)." Did you use a positive control? We recommend OVCAR-3 cell lysate. "As stated in our original query, we used our own positive control, which samples containing total cell lysates prepared from either 1) Xenopus oocytes heterologously expressing human SLC5A8 tagged with V5 (positive) and 2) oocytes injected with an equivalent volume of water (negative control).  Again, as stated in our original query, the first probe was then stripped and re-probed with anti-V5.   Similar findings have been obtained with CHO cells transfected with a similar construct (tagged with 6XHis as well as V5 and probed similarly after stripping).  We also obtained and successfully detected a positive signal using a SLC5A8 antibody from another lab. All told, we performed initial probes with antibodies from the three sources.   "Furthermore, because downregulation of SLC5A8 has been demonstrated in cancer cells, it is unclear to us why OVCAR-3 cells (a cancer cell line!) are considered a good positive control." Do you have an image of the blots with this antibody? "Yes; please see attached pdf.  Samples are total oocyte lysates (i.e. not enriched by biotinylation or immunoprecipitation)." In regard to the choice of positive control for this product. I have spoken with the lab. Of the many cancer and tissue lysates used for quality control of this antibody, the lysate from OVCAR-3 cells gave the best correctly sized band on western blot. I am very sorry that the product did not perform as it should have for you and would like to offer a free of charge replacement (for this product or another) or a credit note/refund which will be applied to the original invoice.
