    why is the actual band size different from the predicted?



    ab27568 detects a band of 34 kDa in MCF7 cells that have been transfected with a vector that overexpresses the SLUG protein. This antibody does not crossreact with the SNAIL protein, as it did not recognize this protein in MCF7 cells overexpressing SNAIL. ab27568 was unable to detect endogenous SLUG in MDA-MB231 cells (not shown).
  • ab27568 staining mouse embryonic stem cells by ICC/IF.  Cells were formaldehyde fixed and permeabilized in 0.1% Triton X-100 prior to blocking with 0.1% BSA and 1% serum for 30 minutes at 21°C.  The primary antibody was diluted 1/100 and incubated for 2 hours at 21°C.  An Alexa Fluor® 488 conjugated goat anti-rabbit antibody, diluted 1/500, was used as the secondary.

I am sorry to hear that a customer is experiencing difficulties with this antibody in IHC. Thank you for passing along the protocol information. Overall this looks pretty standard, and based on how the antibody was tested, should be compatible.


One point of interest is the sample being used by the customer. She indicates she used a rat embryo.Were whole embryos fixed and embedded and then sectioned? What tissue type? How thick were the sections? I am assuming she is not getting any staining at all? Even in her positive control?

La secuencia de aminoácidos del inmunógeno de ab51772 es XXXXXXXXXX . Esta secuencia coincide con la secuencia del inmunógeno del anticuerpo ab27568 que no se encuentra actualmente en stock, por lo que si este anticuerpo os funcionó en el pasado, la alternativa monoclonal debería daros resultados similares.

La secuencia de la proteína es razonablemente conservada en mamíferos, pero no tenemos suficientes referencias para afirmar lo mismo en Oryzias latipes. He intentado llevar a cabo un alineamiento de secuencias pero no he encontrado ni la secuencia de DNA ni la proteica para O. Latines. Es posible que no se encuentre todavía en Genbank (o quizás lo esté con un nombre diferente).
