WB 1/500 - 1/1000. Detects a band of approximately 26 kDa (predicted molecular weight: 29 kDa).
ICC/IF 1/500 - 1/1000.

Target

  • Relevance

    Function: SNAIL is involved in the epithelial to mesenchymal transition (EMT) and formation and maintenance of embryonic mesoderm (By similarity). Binds to 3 E-boxes of the E-cadherin gene promoter and represses its transcription. SLUG is a transcriptional repressor, involved in the generation and migration of neural crest cells. PTM: SNAIL is phosphorylated by GSK3B. Once phosphorylated, it becomes a target for BTRC ubiquitination. Ubiquitinated on Lys-98, Lys-137 and Lys-146 by FBXL14 and BTRC leading to degradation. BTRC-triggered ubiquitination requires previous GSK3B-mediated SNAI1 phosphorylation. Similarity: Both SNAIL and SLUG belong to the snail C2H2-type zinc-finger protein family. Tissue specificity: SNAIL is expressed in a variety of tissues with the highest expression in kidney. Expressed in mesenchymal and epithelial cell lines. SLUG is expressed in placenta and adult heart, pancreas, liver, kidney and skeletal muscle.
  • Cellular localization

    Slug is generally nuclear, while Snail is known to be both cytoplasmic and nuclear. Once phosphorylated (probably on Ser-107, Ser-111, Ser-115 and Ser-119) snail is exported from the nucleus to the cytoplasm where subsequent phosphorylation of the destruction motif and ubiquitination involving BTRC occurs.

    Thank you for your email. I apologize for the delay as I have to leave for a conference.

    I have also received an answer from lab. They have suggested that the recombinant protein they have used is tagged with GST which is why the band size seen in image is 68 kDa rather than 29-30 kDa.

    Looking at the provided image and details, the antibody ab53519 seems detecting the correct target however the protocol might need optimizations in case of ab63568.

    I would suggest following updates for protocol
    - use 5% BSA for antibody dilutions and blocking and block membrane for 1-2 hours at RT.
    - Try no primary control
    - Heat lysates with sample buffer for 10 minutes at 100C.

    Dear whom it may concern,
    Hello. I recently purchased two antibodies from your company, anti-SNAIL and anti-SNAIL+SLUG (phospho S246) to see the phosphorylated form of SNAIL. Since one is detecting SNAIL and the other is detecting phosphorylated form of SNAIL, I was expecting to see bands of similar sizes if not the same, but your data sheets were saying 68 kDa for SNAIL and 26 kDa for phospho-SNAIL. And when I tried the antibodies myself to do Western blotting, my results were not very different: my SNAIL bands did not match with my phospho-SNAIL bands, in fact, I was confused which bands were my phosphorylated SNAIL bands.Why are the sizes markedly different?
