  • IHC image of ab4114 staining in human placenta formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab4114, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

  • ab4114 staining SNAP23 in HT-1080 cells by Immunocytochemistry/ Immunofluorescence.
    Cells were grown on glass coverslips, treated with PMA and then fixed with 4% (w/v) paraformaldehyde in phosphate buffered saline (PBS). Samples were then permeabilized with 0.1% Triton X-100 in PBS and blocked with 5% (w/v) skimmed milk powder in PBS before staining with ab4114 and the appropriate secondary antibody, followed by washing and mounting.

  • Immunohistochemical analysis of rat pancreatic lobules, staining SNAP23 with ab4114. Following fixation in paraformaldehyde, cells were blocked for 30 min at RT, and primary antibody (1/50) was added simultaneously for 1.5 hours at RT. An AlexaFluor®488-conjugated anti-rabbit IgG was used as the secondary antibody.

I'm sorry to inform you have we have no data for the cross reactivity of this anitbody with mouse or other rodent synaptosome associated protein 23. I am not familiar with the degree of sequence homology between human and murine SNAP23. However, as the antibody is a polyclonal it will recognise multiple epitopes across SNAP23 and is therefore more likely to cross react with the protein from different species.
