Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 and blocked with 10% serum for 1 hour at 25°C. Samples were incubated with primary antibody (1/100 in blocking buffer) for 1 hour at 25°C. Thank you for your call today and for your questions about these SNX1 antibodies.
I've heard back from the lab about these antibodies, and unfortunately there are no images available to send, but I did gather some information about the testing.
Ab995 was tested in HeLa cells and the band in Western blot was seen at approximately 70kDa. Ab76072 was tested in immunostaining on mouse cortical cell culture (1 day old) and the WB was done using rat DRG lysate.
Based on this, I think that ab76072 would be much more suitable for your assay. I hope this information will be useful, but please let me know if you have any further questions and I'll be happy to help.
