  • Anti-SNX5 antibody (ab5983) at 1 µg/ml + Jurkat cell lysate (in RIPA buffer) at 35 µg

    Primary incubated for 1 hour. Detected using chemiluminescence.
  • ab5983 at 4µg/ml, staining SNX5 in Human breast tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Steamed antigen retrieval with pH 6 citrate buffer was performed, and the sample was stained with HRP.
  • HeLa cells were grown on coverslips, washed in warm PBS, fixed in warm PFA (3.7% in PBS, 10 minutes), then washed (2x, 5 minutes, PBS), permeabilized (4 minutes, 3% BSA, and 0.5% Triton X-100 in PBS), and blocked in blocking solution (3% BSA in PBS, 30 minutes). Antibody labeling was performed by inversion of coverslips on 20 µl blocking solution with primary or secondary antibodies (1–5 µg/ml).

Thank you for your enquiry and I'm sorry to hear that you are experiencing difficulty with ab5983. At this point I would like to make the following suggestions in order to help improve your results. Decrease the amount of secondary antibody that you are using and also make sure to run a secondary-only control to ensure that these additional bands are not due to your secondary (incubate with the primary only). Decrease the amount of primary antibody that you are using as well. Increase the amount of washes. Make sure that you block with 3% BSA rather than milk. Also, try boiling for 10 minutes rather than 5 minutes. If you need additional assistance, please contact us again.
