    I am sorry to trouble youagain,but I am still wondering the difference of the two products of yours,Anti-SOAT 1 antibody [EP1376Y] (ab53008) and Anti-SOAT 1 antibody (ab39327).Since they detect different bands in Immunoblot analysis,and they have different manufacturing process(method :Synthetic peptide: KMSRLNRLSKSRENPEED, corresponding to amino acids 6/23 of Human ACAT1 and A synthetic peptide corresponding to residues on the C-terminus of human ACAT),i wonder what is the difference between the two when they are used in IHC.

    Answer

    Thank you for contacting us. These two antibodies should work equally well in IHC for Human. ab53008 has not successfully been tested for other species. They do have differenct immunogenic peptides but that doesn't mean one should work better than the other. The WB images both show a band around 50 kD. The additional band you see in ab39327 is detecting the protein in the native form. They would both be guaranteed to work in human for the species lsited, respectively.

    Question

    There is another product of your company Anti-SOAT 1 antibody (ab39327) that detects a band at 47 kDa(Predicted molecular weight: 47 kDa).

    So does it detect a fragment of ACAT-1 or detect the socalled processed form of the protein ?

    Since i didn't find any article that talk about ACAT fragment or processed form,i wonder if i can be informed of more detailed information about your product ACAT-1 antibody(aka SOAT 1).

    Our ab39327 is actually capable of detecting both the native a processed form of the protein. I've attached the WB image and here is the caption:

    Human ACAT-1 has 550 amino acids with an estimated MW of 65 kDa (1). Abcam's affinity purified antibody recognizes a 50 kDa band in most cultured cell lines as reported in the literature (2,3,4). In addition, the antibody reacts with a native form of ACAT-1 at about 65 kDa in liver tissues.
