Target

  • Function

This is the catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with the exchange of sodium and potassium ions across the plasma membrane. This action creates the electrochemical gradient of sodium and potassium ions, providing the energy for active transport of various nutrients.
  • Sequence similarities

Belongs to the cation transport ATPase (P-type) (TC 3.A.3) family. Type IIC subfamily.
  • Post-translational
    modifications

Phosphorylation on Tyr-10 modulates pumping activity.
  • Cellular localization

Cell membrane. Melanosome. Identified by mass spectrometry in melanosome fractions from stage I to stage IV.

Answer

Question

I am interesting in buying an antibody that is specific to Sodium potassium ATPase, that is specific to a peptide of this protein that lies within the cytoplasm. I believe that most commercial ATPase antibodies are specific to regions that are exposed on the cell surface. Do you know whether one of your antibodies actually target the internal face of the plasma membrane?

Asked on Jan 23 2006

Answer

Thank you for your enquiry. The epitope has not been mapped in the cases of ab7671 and ab8344 therefore we do not know which part of the protein is recognised by those antibodies. As the immunogen used to generate ab353 is purified Na,K-ATPase from rabbit renal outermedulla and the antibody is a polyclonal antibody the product will recognise a number of epitopes on the protein, some are likely to be at the plasma membrane portion intracellularly and some extracellularly. I hope this information helps, please do not hesitate to contact us again if you need more information,

Asked on Dec 18 2001

Answer

This antibody was found to be reactive against the intact enzyme and enzyme subunits (alpha at 100 kDa and beta at 50 kDa) as well as glycosylated and non-glycosylated enzyme forms by western blot. The product is also reactive against 1.0ug of ATPase, Sodium, Potassium in a standard sandwich ELISA.

Answer

Reactivity with other mammalian sources is unknown, but likely, due to the nature of the polyclonal antibody.
