Application Abreviews Notes
WB Use a concentration of 1 µg/ml. Detects a band of approximately 40 kDa (predicted molecular weight: 39 kDa).
ICC/IF Use at an assay dependent concentration.
IHC-P 1/500 - 1/1000. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

Images

  • ICC/IF image of ab87775 stained human embryonic stem cell derived neural progenitor cells. The cells were fixed in 4% Paraformaldehyde for 15 minutes at RT, before incubation in 0.3% Triton x-100 to permeabilize the cells and blocking with 10% serum to minimize non-specific protein-protein interactions. The cells were then incubated with Sox1 antibody (ab87775, 1/500 dilution) for one hour at 20°C in 0.3%Triton x100, 10%Goat serum, 2.5% BSA. The secondary antibody (green) was an Alexa Fluor® 488 goat anti-rabbit IgG used at a 1/500 dilution.

    Exposure time: 8 minutes


    This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% BSA before being incubated with ab87775 overnight at 4°C. Antibody binding was detected using an anti-rabbit HRP secondary antibody, and visualised using ECL development solution ab133406.
