Opossum (Monodelphis domestica) primary dissociated cortical cells from postnatal day (P) 6 pups were fixed with 4% paraformadehyde, incubated with 0.1 M glycine, permeabilized with 0,1% Triton X-100 and saturated with 0,5% bovine serum albumin (BSA) (all solutions made in PBS). Primary anti-SOX2 antibody [9-9-3] (ab79351) (1:200 dilution) was incubated for 1h at room temperature (RT). Secondary antibody (goat anti-mouse Alexa Fluor 488, 1:500 dilution in PBS) was used (incubation time 30min at RT). Nuclei were stained with 2 µg/ml Hoechst 33342.
