  • IHC (PFA-fixed) image of SOX6 (ab30455) on sections of P18 mouse dentate gyrus. The mice were perfusion fixed with 4% PFA and then post-fixed in 4% PFA for a few hours. The blocks were prepared in agarose/sucrose and cut using a vibratome (50 µm). The sections were permeabilized with 1% Triton X-100 for 30 mins and blocked with 10% FBS for 2 hours at room temperature, then incubated overnight at 4°C with the primary antibody. The primary antibody was used at a 1/100 dilution and detected with donkey anti-rabbit 488 secondary at 1/1000 dilution.

  • ab30455 staining Sox6 in post-natal mouse brain sections. PFA-fixed post-natal mouse brain sections were incubated for 30 minutes in blocking solution followed by an overnight incubation with ab30455 (1/500) at 4°C. No staining was observed in sections obtained from Sox6 knockout mice.
  • IHC-FoFr image of Sox6 staining in Rat hippocampus sections using ab30455.The sections used came from animals perfused fixed with Paraformaldehyde 4%, in phosphate buffer 0.2M. Following postfixation in the same fixative overnight, the brains were cryoprotected in sucrose 30% overnight. immunostainings were performed using the ‘free floating’ technique.

- Could you provide the probable date when the previous batch was purchased? Is it last month, if yes then that batch and the current batch is same.

LOT NUMBER 958128 ORDER NUMBER 2011/4563 DESCRIPTION OF THE PROBLEM this antibody gives me lot of background. My collaborators reordered the same reference (unfortunately, the same lot). I tested it also and got the same absence of specific staining. SAMPLE E10.5 mouse embryo section PRIMARY ANTIBODY Anti Sox6 abcam ab30455 (polyclonal from rabbit): I tried several dilutions, inlcuding the recommended one 1/400, diluted in the blocking solution and incubated in a humidified chamber over night at 4°C. I washed the next day 3 times in PBS or PBS-Tween20 0.1% at least 10 minutes, 3 times DETECTION METHOD IF confocal microscopy POSITIVE AND NEGATIVE CONTROLS USED I am working with embryo sections. I know that Sox6 is expressed on my samples because I did IF with another antibody and got very nice signal as expected in the ventral part of the neural tube. Unfortunately, I do not have this antibody anymore and I had to find another one... ANTIBODY STORAGE CONDITIONS the antibody is stored at 4°C and was shipped already as liquid FIXATION OF SAMPLE 4%PFA 60 minutes 4°C or 0.2% PFA over night 4°C, embedded in gelatin/sucrose and cryosectionned ANTIGEN RETRIEVAL I tried without or with the antigen retrieval kit from Dako PERMEABILIZATION STEP I tried with or without permeabilization 20' in PBS-Triton X100 0.05% BLOCKING CONDITIONS I tried different blocking solution: FCS 5% in PBS Tween 20 0.1% or BSA 3% - goat serum 1% in PBS Tween 20 0.1% both 20' at least at RT SECONDARY ANTIBODY I used different Alexa from invitrogen coupled with 488 or 546 fluorochromes and made in different species. The background is not due to the secondary antibodies since I have tried several ones and I am using these secondary antibodies without any problems HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 10 HAVE YOU RUN A "NO PRIMARY" CONTROL? Yes DO YOU OBTAIN THE SAME RESULTS EVERY TIME? Yes WHAT STEPS HAVE YOU ALTERED? It would be very helpful for our quality investigation if you could please provide an image of your staining. You may be able to reduce the background staining by blocking longer (up to overnight at 4C) and using 10% normal serum from the host species of your secondary antibody as the blocking agent. If your signal is strong, it may also help to reduce the primary antibody incubation time to 1-4 hours at room temperature. If you are unable to improve your results, please let me know and I would be happy to offer you a free of charge replacement with one of our other SOX6 antibodies, such as ab66316 or a credit or refund.
