The results show multiple bands; therefore this customer wants to ask for your help to modify his experiment step, could you please offer any suggestion to improve his wb result? I attached the wb image in this letter and his experiment step as follow:   1. After reading through the detailed protocol you kindly forwarded to Abcam, I would like to make the following comments/suggestions: 1) It would be worth preparing nuclear lysate and run on the gel - instead of total cell lysate. 2) Please advise your customer to use positive control (purified recombinant SPANX-C). 3)  Does the detection system work fine? Have you used it successfully with another primary antibody? Have you run a no primary - only secondary antibody - control to see if any of the non-specific bands are due to the secondary or not? If you have not done yet, I would advise you to check it.   I hope this will be useful for you. Thank you for your enquiry. I have investigated this with the originator who provided this image. All the samples in lane 2 to 5 of this western blot were recombinant fusion proteins. Consequently, the molecular weights are much higher than expected for the endogenous protein. I hope this is helpful to you.
