Overview

  • Product name
    Anti-Spermine antibody
  • Description
    Rabbit polyclonal to Spermine
  • Host species
    Rabbit
  • Specificity
This antibody reacts with spermine. Cross-reacts with spermidine 100%.
  • Tested applications
Suitable for: ICC/IF, ChIP, WBmore details
  • Immunogen

    Spermine conjugated to KLH.

  • General notes

Polyamines are substrates for transglutaminase enzymes and can be covalently conjugated to proteins. In this case Western Blot and other methods can be able to detect polyaminated proteins.

Properties

Applications

Our Abpromise guarantee covers the use of ab26975 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use at an assay dependent concentration.
ChIP Use at an assay dependent concentration.
WB 1/1200 - 1/5000.

Target

  • Relevance
Spermine, a polyamine, is a positively charged compound found in living cells and can cause conformational changes in DNA via charge and solubility mechanisms which can affect gene expression cell differentiation. Spermine may be linked to the development of cancer.
  • Alternative names
    • Spermine antibody

Images

  • ab26975 staining spermine in the Human HeLa cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with 4% formaldehyde for 15 minuts at room temperature (Upper images) or iced methanol of 3 minutes (Lower images) and blocked with 2% BSA for 30 minutes at 27°C. Samples were incubated with primary antibody (1/100) for 1 hour at 27°C. An Alexa Fluor®568-conjugated Donkey anti-rabbit polyclonal (1/1000) was used as the secondary antibody. Samples handled without ab26975 were used as negative controls (Left-hand panels).

Answer

Thank you for contacting us.

Spermidine and spermine are both polyamines found in living tissues and have various metabolic functions. As they are not proteins, they are not species specific. If the tissue you are interested in probing has spermine or speridine present the antibodies ab7318 and ab26975 should be able to detect the molecules.

I hope this information has been of help. If you require any further information please do not hesitate to ask.

Answer

Thank you for contacting us and your interest in our products.

The two antibodies which you are interested in (ab7318 and ab26975) are both formulations of the whole antiserum. This means that quantification of the exact concentration is not possible as there are several other proteins as well as the active antibody within the preparation. Typically, whole antiserum is of a concentration of 1-10 mg/ml.

I would suggest optimising the dilution of the antibodies for your experiment, starting with a dilution of 1/50-1/100. From there you should be able to see if you need to reduce or can increase the dilution needed for the optimum results. Ab26975 has not as yet been tested for its use in immunohisotchemistry, we would therefore be very interested in the results you obtain with this antibody. By submitting an abreview for these antibodies you would recieve Abpoints which can be redeemed against the purchase of products from our catalogue, or amazon vouchers.
