      Splice factor that binds to the single stranded 3'AG at the exon/intron border and promotes its utilization in the second catalytic step. Involved in the regulation of alternative splicing and the utilization of cryptic splice sites. Promotes the utilization of a cryptic splice site created by the beta-110 mutation in the HBB gene. The resulting frameshift leads to sickle cell anemia.

    Answer

    The antibody has not been tested on mouse samples before, so there may be an issue with low cross-reactivity with mouse RBM17, though reactivity is predicted based on homology with the immunogen. In any case, the high background is the main issue.

    If the new lysate you are preparing, and/or the switch we discussed to milk from BSA for blocking/diluting does not help, you may want to consider preparing samples from isolated nuclei, to enrich for the protein. Detection may be difficult with any antibody against this target, if the protein is scarce. Another approach is to immunoprecipitate it, for instance with the antibody I mentioned, ab114968, though it has not been tested against mouse protein either.

    Re your RBM17 antibody (#ab101441), can you tell me why it picks up an ˜70kda band in addition to the ˜54 and 45kda bands (which correspond with its molecular weight)?

    In my opinion, the 54 kDa band is phosphorylated RBM17 and the lower 45 kDa band is the dephosphorylated version. The 70 kDa band appears to be a non-specific band.
