    Receptor for interleukin-33 (IL-33), its stimulation recruits MYD88, IRAK1, IRAK4, and TRAF6, followed by phosphorylation of MAPK3/ERK1 and/or MAPK1/ERK2, MAPK14, and MAPK8. Possibly involved in helper T-cell function.

    Transfer condition: Wet, 250mA, 2 hrs (NC membrane).

    Blocking condition: 5% non-fat milk in TBS at 4°C overnight. 

    Washing condition: 5ml TBST, 4 x 5 min. 

    Exposure system: GE ECL 1 min. 

    Target bands were observed at 70kD in Human, mouse and rat cells and at 39kD in mouse cells.

Answer

For staining formalin-fixed, paraffin-embedded tissue with the ST2 antibody ab25877, we recommend a heat-mediated antigen retrieval with 10mM sodium citrate, pH 5.5, at 95-100C. The optimal time of incubation will need to be determined for your samples but 20 minutes is typically sufficient.

> 130 kDa
˜ 100 kDa
˜ 70 kDa
˜ 40 kDa
Les masses moléculaires théoriques indiquées dans UniProt (chez la souris) sont de 38.5 kDa pour la forme soluble sST2 et de 64.8 kDa pour la forme membranaire ST2L. Nous voudrions voir s'il y a une différence d'expression entre les 2 formes (soluble et membranaire) mais vu le pattern de bandes obtenu nous avons des doutes sur la spécificité de cet anticorps. Par ailleurs, il est difficile de trouver les tailles exactes des bandes montrées dans les articles.

Pourriez-vous SVP nous dire si l'anticorps est bien spécifique et si oui à quoi correspondent les différentes bandes observées? Auriez-vous SVP des références à nous indiquer?

Merci d'avoir pris le temps de remplir notre questionnaire et de nous avoir contactés. Je suis désolée d'apprendre que vous avez eu des difficultés à obtenir de bons résultats avec ab25877

Notre politique est d'offrir tout d'abord la meilleure assistance technique. Si nous concluons que le produit que vous avez reçu ne fonctionne pas comme mentionné sur sa fiche technique et que ce produit a été commandé sous 120 jours, nous nous ferons un plaisir de vous offrir un remplacement gratuit ou un avoir en compensation.

En regardant vos résultats de western blot, je peux voir que la bande la plus importante est celle qui représente isoform B. Cette bande devrait être à 38.5 kDa mais semble légèrement décalé et apparait plus haut vers 40 kDa. Il me semble que toutes les bandes ont migré légèrement vers le haut ce qui expliquerait pourquoi la bande qui représente l'isoforme A apparaît à 70 kDa, un peut plus haut que prévue (64.8 kDa.)

Par contre c’est vrai que les deux bandes à 100 kDa et 130 kDa ne devraient pas y être. J'aimerais donc vous offrir quelques suggestions afin d'optimiser vos résultats.

Lors de la préparation de vos échantillons, je vous suggère d'augmenter le temps d'ébullition de 5 minutes à 10 minutes et d’utiliser un « sample buffer » frais. Ceci est pour éviter que des multimères se forme puisque la bande à 100 kDa pourrait correspondre à un dimère de l'isoforme B et la bande à 130 kDa pourrait correspondre à un dimère de l'isoforme A ou un trimère de l'isoforme B.
Essayez de changer l'agent bloquant à 5% BSA. Certains anticorps donnent des résultats plus spécifiques en utilisant de la BSA plutôt que de lait en tant qu’agent bloquant. Voir l’exemple d’un western blot pour Abcam anti-GAPDH (ab9385): https://www.abcam.com/ab9385.
Changer la concentration de l’anticorps. Notre fiche technique suggère d’utiliser une concentration de 1-2 ug/ml en Western blot. Je vous recommande donc d’utiliser une dilution de 1:1000 plutôt que 1 :300 pour réduire les bandes non-spécifiques.

Je suis désolée d'apprendre qu’ab25877 ne vous donne pas de bons résultats en Western blot.

L’anticorps ab25877 devrait effectivement reconnaitre les 2 isoformes de la protéine à 38.5 kDa et 64.8 kDa ce qui expliquerait les bandes observées à 70 kDa et 40 kDa. J’ai fais des recherches de mon côté et je n’ai pas trouvé des explications pour les bandes à 100kDa et > 130kDa. Pour mieux comprendre ce cas, veuillez trouver ci-joint un questionnaire qui nous permettra de regrouper le plus d'informations possible sur le protocole que vous avez utilisé avec cet anticorps.
Celui-ci nous permettra de vous fournir la meilleure assistance technique.

Si nous concluons que notre produit est fautif et ne fonctionne pas comme explicité par notre fiche technique, nous serons ravis de mettre en place l'envoi d'un tube de remplacement gratuit ou d'un remboursement (sous réserve d'une commande ultérieure de 120 jours).

We only have one antibody which has been tested (and would be covered by the Abpromise guarantee) for IHC. This is the rabbit polyclonal https://www.abcam.com/ST2-antibody-ab25877.html. This has been raised against an immunogen taken from the mouse protein. We have not tested this antibody with the human protein. However, the immunogen used shares 81% homology with the human protein. It is therefore possible that the antibody may be able to detect the human protein however, we would not be able to guarantee this.

Alternatively we have an antibody which has been used to detect the mouse and human protein in western blotting but we have not used it in IHC. This is the rabbit polyclonalhttps://www.abcam.com/ST2-antibody-ab72778.html. Again, as we have not tested this ourselves in IHC-P we cannot guarantee that it would work.
