Target

  • Relevance
    S.aureus binds IgG molecules by their Fc region. In serum, the bacteria will bind IgG molecules in the wrong orientation on their surface, which hinders opsonization and phagocytosis. Mutants of S. aureus lacking protein A are more efficiently phagocytosed in vitro, and mutants in infection models have diminished virulence. Due to its affinity for the Fc region of many mammalian immunoglobulins, protein A is considered a universal reagent in biochemistry and immunology.

Ab20002 - Mouse monoclonal [702] to Staphylococcus aureus and Ab37644 - Mouse monoclonal [704] to Staphylococcus aureus recognize peptidoglycan which is a cell wall component, so, by definition, this is on the surface of the cells.

About the sample type, we are sorry that we have not tested ab20002 and ab37644 for detecting Staphylococcus aureus in blood sample, so we do not have data to support whether these items will work well or not in regarding to the detection of Staphylococcus aureus in blood sample.

I havecontacted the source for all the Staphylococcus aureus antibodies we have that have been tested in WB and/or ELISA to ask if they have been tested for crossreactivity. Here is the information:

ab20002 Anti-Staphylococcus aureus antibody [702]

This antibody specifically recognizes peptidoglycan of Staphyloccus aureus, Protein A-negative Staph. aureus and Staph. epidermidis. It does not cross-react with Streptococcus sp., Mycoplasma sp. or Corynebacteria sp.However, as far as I know it has not been tested with another Staphlococcus strains.

ab21028 Anti-Staphylococcus aureus antibody [702] (Biotin)

This is the biotinylated version of ab20002 with the same specificity and cross-reactivity data.

ab37644 Anti-Staphylococcus aureus antibody [704]

This antibody reacts with Staphyloccus aureus, Protein A-negative Staph. aureus and Staph. epidermidis. Does not cross-react with Corynebacteria sp., Streptococcus sp. or Mycoplasma sp. Again, as far as I know it has not been tested with another Staphlococcus strains.

ab68952 Anti-Staphylococcus aureus antibody (HRP)

The only other strain of Staphylococcus that this antibody has been tested is Staph. albus, which does cross-react. However, we believe that it will probablycross-react with other strains of Staph. bacterium because the full bacteria proteome is used as the immunogen.

ab73962 Anti-Staphylococcus aureus antibody

Reactivity of this antibody is confirmed for Staphylococcus aureus. We do not know about reactivity of other strains.

I am sorry we regrettably do not have an antibody from the catalog that we can confirm will definitely not detect other strains of Staphylococcus. However, I hope this is helpful. If you have any further questions, please do not hesitate to contact me.

To be able to help you further in this matter, could you please send me a copy of the protocol that you have been using with ab37644, including any variations that you have tried to achieve better staining. Information that would be useful to us is, sample types, fixations, permeabilization steps, primary antibody concentrations and incubations times etc. Also when you say "not been able to get a good signal from the antibody", what does that mean? Are you seeing the correct staining pattern but its very weak or do you just get high background?

If we cannot resolve the issue that you are having, then I would be happy to process a refund for you. To do that could you also include the Abcam order number ort the PO# number that was used to purchase this antibody.

The antibody ab37644 recognizes an epitope on the peptidoglycan. The epitope recognized is not on Protein A.
