  • Immunofluorescence analysis of Potorous tridactylis kidney (PtK2) cells, staining Streptavidin with ab10020.

    Cells were fixed with 4% paraformaldehyde, before being incubated with wt streptavidin (2 µM) for 5 min at room temperature. Samples were incubated with 1 µL primary antibody for 1 hour at 4°C. A FITC-conjugated goat anti-mouse IgG was used as the secondary antibody.

Answer

Normally 10-50 ug of lysate protein is required for an antibody detection in western blot, however if you want to detect amount lower than this then you may need to isolate the protein or purify the protein form the cell lysates. e.g. antibody can easily detect 10-100 ng of purified protein.

Secondly you would need a strong detecting reagents e.g. https://www.abcam.com/optiblot-ecl-max-detect-kit-46pg-47ng-ab133408.html which can detect the antibody dilution 1/5000 to 1/20,000 with protein concentration in picogram.

I think any of the any of the antibodies you are interested in would work for you in your desired applications. As they are mouse monoclonals, you may see some mouse on mouse staining and unfortunately we do not have information as to which would give the least cross reactivity.

Wie versprochen habe ich genauer in den Laboren nachgefragt, die unsere Streptavidin-Antikörper herstellen, und dabei hat sich herausgestellt, dass unsere monoklonalen anti-Streptavidin-Antikörper (ab10020-10023), die Sie angesprochen hatten, jeweils das gesamte Streptavidin erkennen - die Epitope wurden unseres Wissens leider nicht kartiert - und hauptsächlich für die Amplifikation von Signalen in verschiedenen Anwendungen wie Immunhistochemie verwendet werden.

Wenn ich Sie richtig verstanden hatte, sind Sie jedoch an einem Antikörper gegen ein C-terminales Strep-tag interessiert, was ja vom oben implizierten Biotin/Streptavidin-System verschieden ist. Gegen dieses Strep-tag hätten wir ab76949 im Katalog, der das Strep-tag der angegebenen Sequenz unabhängig davon erkennt, ob es C- oder N-terminal lokalisiert ist:

Wir können garantieren, dass dieser Antikörper im Western blot funktioniert (www.abcam.com/Abpromise). I can confirm that ab59320 is now unpublished and not available any more. I am sorry if this causes any inconvenience.

ab6676 is a polyclonal antibody that has the whole streptavidin molecule as immunogen. It was not tested if the binding sites of biotin are blocked, but we expect this due to the polyclonal nature of this antibody. This is not part of the Abpromise guarantee.

ab10022 reacts with the target protein at a non-overlapping epitope with ab10023, ab10020 and ab10021 epitopes. However, we have not determined the exact epitopes for these antibodies. We are confident all these antibodies bind to streptavidin without interfering with the biotin-binding reaction.
