IHC-P Use a concentration of 0.1 - 0.5 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

  • Function
    Exhibits arylsulfatase activity and highly specific endoglucosamine-6-sulfatase activity. It can remove sulfate from the C-6 position of glucosamine within specific subregions of intact heparin. Diminishes HSPG (heparan sulfate proteoglycans) sulfation, inhibits signaling by heparin-dependent growth factors, diminishes proliferation, and facilitates apoptosis in response to exogenous stimulation.
  • Tissue specificity
    Expressed at highest levels in testis, stomach, skeletal muscle, lung, kidney, pancreas, small intestine and colon. It is also detected in normal ovarian surface epithelial cells. Down-regulation seen in ovarian carcinoma cell lines, ovarian cancers, breast, pancreatic, renal and hepatocellular carcinoma cell lines.

  • IHC image of Sulfatase 1 staining in Human skeletal muscle formalin-fixed paraffin embedded tissue section*, performed on a Leica Bond™ system.. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab32763, 0.1 µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. 

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Thank you for contacting us yesterday and for sharing the problems you have been experiencing with the Anti-Sulfatase 1 antibody (ab32763).
This antibody is a Fast Tranck antibody and as such has as yet not been tested in anything other than ELISA against its immunising peptide in our hands and we are therefore not able to guarantee its performance. This information in stated on the from of the datasheet of ab32763. We have however received some reports of customers having used it successfully in both Western blotting and IHC. Question

J'ai commandé en mai 2012 un anticorps anti-sulfatase 1 (ab32763) (devis n°41036), et nous l'avons utilisé en western blot pour détecter la protéine dans des cellules fibroblastes. Dans ces cellules, le transcript de la sulfatase est détecté en RT-PCR en temps réel avec des Ct (seuil de détection) proches de 20, ce qui nous suggère que la protéine y est présente en bonne quantité et devrait facilement être détectable par immunorévélation.
Nous avons donc acheté votre anticorps anti-sulfatase pour détecter la protéine cellulaire. Après dépot de 60µg de protéines totales sur un gel en SDS-PAGE et transfert sur nitrocellulose, nous avons utilisé l'anti-sulfatase 1 comme anticorps primaire et le résultats est non satisfaisant. Plusieurs conditions ont été essayées (tampon d'incubation, durée d'incubation, dilution).
Par contre, les mêmes échantillons ont également été testés pour détecter la protéine 6OST1 avec votre anticorps anti-HS6ST1 (ab88488).
Dans ce cas-çi, les résultats sont interprétables et nous satisfont.
Je vous joint en attaché un fichier pdf avec nos résultats obtenus avec l'anticorps anti-sulfatase1, qui cause problème ! et l'anticorps anti-HS6ST1, qui nous convient (témoin de notre bonne foi)....Théoriquement nous attendions une augmentation de la sulfatase 1 en réponse au stimulus étant donné qu'en RT-PCR, le transcrit est augmenté x 10...de plus, l'invalidation par des siRNA spécifiques de la sulfatase (validés en RT-PCR en temps réel) ne provoque aucune diminution de la protéine détéctée en western blot...Ce qui n'est pas satisfaisant...De plus, comme vous pouvez le voir, aucune bande n'est spécifique de la sulfatase1, qui devrait être soit > 100kDa (pour la forme pro-enzyme), soit environ à 75kDa (pour la forme mature).
Abcam propose un remplacement ou un remboursement des produits qui ne satisfont pas. Est-il donc envisageable de remplacer l'ab32763 par l'autre anticorps anti-sulfatase 1 que vous distribuez ? La référence de ce nouveau anticorps est le N°ab92300.

The protocol seems fine however there is only one suggestion we would like to offer; heating the lysates for 10 minutes at 100C may improve the results.

This protein is very heavily glycosylated so the observed band size will be higher than the predicted one. I would suggest searching literature for the observed band size.

Please also note that this is a fast track antibody. We do not have full characterization data for this antibody which is why it is much cheaper. The Abcam money back or replacement guarantee will not be applied to this product; the full terms and conditions are give on the datasheet.
